Pilot fase I/II undersøgelse af aminosyreleucin i behandling af patienter med transfusionsafhængig Diamond Blackfan Anæmi (LeucineDBA)
Brugen af nye terapier til at rekonstituere blodcelleproduktion og fremme organydelse ved brug af knoglemarvssvigt som model: en pilotfase I/II undersøgelse af aminosyreleucin i behandlingen af patienter med transfusionsafhængig Diamond Blackfan Anæmi
Denne undersøgelse vil bestemme sikkerheden og muligheden for at give aminosyren leucin til patienter med Diamond Blackfan anæmi (DBA), som er afhængige af transfusioner af røde blodlegemer.
Leucinen forventes at give et respons hos patienter med DBA til det punkt, hvor produktionen af røde blodlegemer øges. Transfusioner af røde blodlegemer kan derefter være mindre hyppige eller muligvis afbrydes.
Efterforskerne vil undersøge eventuelle bivirkninger ved at give leucin til DBA-patienter. Leucinniveauer af leucin vil blive opnået ved baseline og under undersøgelsen.
Lægemidlet leucin leveres i kapselform og tages 3 gange dagligt i i alt 9 måneder.

Leucin vil blive givet til deltagerne i form af en kapsel og vil blive taget tre gange dagligt.
Blodhæmoglobinniveauet vil blive overvåget hver 3.-4. uge i 9 måneder.
Hele undersøgelsen varer 12-15 måneder.
Forsøgspersoner skal være to år gamle eller ældre og have transfusion i mere end seks måneder før tilmelding.

Inklusionskriterier:
- diagnosticeret med Diamond Blackfan anæmi som offentliggjort i British Journal of Hematology
- transfusionsafhængig
- alder 2 år og ældre
- tilstrækkelig nyrefunktion
- tilstrækkelig leverfunktion
- negativ B-HCG, hvis patienten er en menstruerende kvinde og dokumentation for tilstrækkelig prævention
- underskrevet informeret samtykke
Ekskluderingskriterier:
- Kendt overfølsomhed over for forgrenede aminosyrer
- Diagnose af en medfødt fejl i aminosyremetabolismeforstyrrelser
- Forudgående hæmatopoietisk stamcelletransplantation
- Graviditet, eller planlægger at blive gravid under forsøgets varighed
