Badanie bezpieczeństwa szczepionki pomagającej chronić zdrowe dzieci przed boreliozą
FAZA 3, RANDOMIZOWANA, KONTROLOWANA PLACEBO, BADANA Z ZAŚLEPIONĄ DLA OBSERWATORÓW, ABY OCENIĆ BEZPIECZEŃSTWO 6-WALENTNEJ SZCZEPIONKI PRZECIWKO BIORILIIE OSPA BAZUJĄCEJ (VLA15) U ZDROWYCH DZIECI W WIEKU OD 5 DO 17 LAT
Celem tego badania jest ustalenie, czy badana szczepionka (zwana VLA15) jest bezpieczna dla zdrowych dzieci.
Poszukujemy dzieci, które:
- są zdrowe
- są w wieku od 5 do 17 lat
- u których w przeszłości nie zdiagnozowano żadnej formy boreliozy
- nie otrzymało w przeszłości żadnych szczepionek przeciwko boreliozie
Borelioza występuje najczęściej u dzieci w tym wieku. Badana szczepionka może potencjalnie pomóc w zapobieganiu boreliozie. Celem tego badania jest uzyskanie większej ilości informacji na temat bezpieczeństwa badanej szczepionki w tej grupie wiekowej.
Uczestnicy będą w tym badaniu przez około 2 lata. W tym czasie otrzymają VLA15 lub placebo (sterylny roztwór słonej wody) poprzez „zastrzyk” w ramię. Porównamy doświadczenia dzieci otrzymujących VLA15 z doświadczeniami dzieci otrzymujących placebo. Uczestnicy nie będą wiedzieć, czy otrzymują VLA15, czy placebo.
Każdy, kto weźmie udział w tym badaniu:
- dostać strzały w klinice lub w gabinecie szpitalnym
- otrzymać łącznie 4 strzały
- otrzymać pierwsze 3 strzały w ciągu 6 miesięcy
- otrzymać ostatni zastrzyk około 1 rok później
- konieczność przybycia do ośrodka badawczego na 6 zaplanowanych wizyt; 4 z nich to wizyty szczepień, a 2 to wizyty kontrolne. W trakcie badania skontaktujemy się z Tobą telefonicznie 1 raz w roku, aby monitorować Twoje doświadczenia. Możesz mieć dodatkowe wizyty, jeśli wystąpi ciężka reakcja po dawce szczepionki.

Opis
Kryteria przyjęcia:
- Zdrowi uczestnicy w momencie rejestracji, którzy zostali uznani za kwalifikujących się do włączenia do badania. Uczestnicy z istniejącymi wcześniej przewlekłymi schorzeniami, które zostały uznane za stabilne, mogą zostać uwzględnieni.
- Uczestnicy i/lub rodzice/opiekunowie uczestników, którzy chcą i są w stanie przestrzegać wszystkich zaplanowanych wizyt, procedur badania i warunków związanych ze stylem życia w czasie trwania badania.
Kryteria wyłączenia:
- Uczestniczki, które są w ciąży, karmią piersią lub mają pozytywny wynik testu ciążowego z moczu podczas Wizyty 1. Aktywne seksualnie kobiety i płodni mężczyźni, którzy nie chcą stosować antykoncepcji zgodnie z protokołem.
- Wszelkie przeciwwskazania do szczepienia lub składników szczepionki, w tym wcześniejsza reakcja anafilaktyczna na jakąkolwiek szczepionkę lub składniki szczepionki.
- Jakakolwiek diagnoza boreliozy w ciągu ostatnich 3 miesięcy.
- Jakakolwiek historia boreliozy z Lyme, zapalenia serca, neuroboreliozy lub innej rozsianej boreliozy (LD), niezależnie od tego, kiedy została zdiagnozowana.
- Znane ugryzienie przez kleszcza w ciągu ostatnich 4 tygodni.
- Wrodzony lub nabyty niedobór odporności lub inne stany lub terapie związane z immunosupresją, które hamują zdolność do wywołania odpowiedzi immunologicznej na szczepionkę.
- Inny stan medyczny, psychiatryczny, aktywne myśli/zachowania samobójcze lub nieprawidłowości laboratoryjne, które zwiększają ryzyko udziału w badaniu lub, w ocenie badacza, są nieodpowiednie do badania.
- Pokwitowanie poprzedniego szczepienia przeciwko LD.
- Leczenie LD w ciągu 3 miesięcy przed podaniem interwencji w ramach badania.
- Otrzymanie produktów z krwi/osocza lub immunoglobulin w ciągu 6 miesięcy przed podaniem interwencji w ramach badania do zakończenia badania.
- Przyjmowanie ogólnoustrojowych kortykosteroidów przez ≥14 dni w ciągu 28 dni przed podaniem interwencji w ramach badania. Dozwolone są kortykosteroidy wziewne/nebulizowane, dostawowe, dokaletkowe lub miejscowe.
- Otrzymanie przewlekłego leczenia ogólnoustrojowego innymi znanymi lekami immunosupresyjnymi lub radioterapią w ciągu 6 miesięcy przed podaniem interwencji w ramach badania.
- Bieżące stosowanie jakichkolwiek zabronionych leków towarzyszących lub uczestnicy, którzy nie chcą/nie mogą stosować dozwolonych leków towarzyszących.
- Uczestnictwo w innych badaniach obejmujących badane leki/szczepionki/urządzenia w ciągu 28 dni przed rozpoczęciem badania i/lub w trakcie udziału w badaniu (dopuszczalne są badania obserwacyjne).
- Personel ośrodka badawczego, delegaci sponsorów/sponsorów bezpośrednio zaangażowani w prowadzenie badania oraz członkowie ich rodzin; personel ośrodka nadzorowany przez badacza i członków ich rodzin.

An SAE was defined as any untoward medical occurrence that, at any dose, met one or more of the following criteria: resulted in death, was life-threatening, required inpatient hospitalization or prolongation of existing hospitalization, resulted in persistent or significant disability/incapacity, was a congenital anomaly/birth defect, any other important medical event.
