详细说明
诊断为浸润性乳腺癌的 50 岁以上女性全部符合:淋巴结阴性/N0i+; T1 或 T2;低级或中级;激素受体(ER 和 PR)的强表达或中等表达;和 HER2 阴性,并且已经对其肿瘤进行了充分的局部治疗,被邀请参加 Prosigna® 筛查。
他们切除的肿瘤样本被送去进行 Prosigna® 检测。

为了模拟现实生活,如果出现无法忍受的副作用,可以在两年内灵活地切换到另一种内分泌治疗标准。
在两年的治疗期间,参与者每六个月接受一次检查,并完成定期的生活质量 (QOL) 调查问卷,旨在衡量生活质量、情绪、副作用和焦虑/对复发的恐惧。
经过两年的治疗,参与者每年进行一次研究访问,直到第 10 年,在某些但不是所有时间点进行类似的问卷调查,并对研究终点进行评估。
主要终点是 5 年无远处复发间隔 (DRFI),定义为 5 年内无远处复发或乳腺癌死亡。
次要终点包括纵向 QOL 比较、10 年无乳腺癌间隔和 10 年对侧乳腺癌发生率。
预计需要筛选约 400 人才能招募 290 名具有必要低分的参与者。
样本量基于 96.8% 的预期 5 年 DRFI、0.05 的单侧 alpha 以及如果单侧置信区间的下边界产生低于 95% 的观察到的 DRFI 则拒绝备选假设。

- 单焦点或多焦点(不是多中心)
- 单方面
- 中度或强激素受体阳性
- HER2阴性
- 1 级或 2 级导管,或任何级别的小叶状,或纯管状(任何级别)或纯乳头状(任何级别)。 如果混合性小叶-导管组织学,导管成分必须为 1 级或 2 级。
- 分期 pT1N0(肿瘤 </= 20mm,淋巴结阴性)或 pT2N0(肿瘤 21-50mm,淋巴结阴性)或 pT1N0i+(肿瘤 </=20mm 且淋巴结[s]中的孤立肿瘤细胞)或 pT2N0i+(肿瘤 21-50mm 且孤立节点[s]中的肿瘤细胞)(见附录2)。 肿瘤大小必须足以进行 Prosigna® 测试。 pNX(节点状态未知)阶段不符合条件。
