En 6-måneders sikkerheds- og fordelsundersøgelse af inhaleret fluticasonpropionat/salmeterol kombination versus inhaleret fluticasonpropionat til behandling af 6.200 pædiatriske forsøgspersoner 4-11 år gamle med vedvarende astma

Formålet med denne undersøgelse er at vurdere, om risikoen for alvorlige astma-relaterede hændelser (astma-relaterede hospitalsindlæggelser, endotracheale intubationer og dødsfald) hos børn 4-11 år gamle, der tager inhaleret fluticasonpropionat/salmeterol-kombination, er den samme som dem, der tager inhaleret fluticasonpropionat alene.

En historie med klinisk skoldkoppeinfektion eller modtager af en skoldkoppevaccine i lande, hvor produktetiketten indeholder en advarsel om mere alvorlige skoldkoppeinfektioner hos patienter, der bruger kortikosteroider.

Historie om livstruende astma
Ustabil astma
Nuværende brug af højdosis ICS- eller ICS/LABA-terapi til behandling af astmasymptomer
Samtidig luftvejssygdom: Aktuelle tegn på lungebetændelse, pneumothorax, atelektase, lungefibrotisk sygdom, allergisk bronkopulmonal aspergillose, cystisk fibrose, bronkopulmonal dysplasi eller andre alvorlige respiratoriske abnormiteter bortset fra astma.
Luftvejsinfektion
Forsøgspersoner med kun anstrengelsesudløst astma
En astmaforværring inden for de sidste 4 uger eller mere end 4 separate eksacerbationer inden for de sidste 12 måneder
Indlæggelse for astma inden for 4 uger eller mere end 2 indlæggelser inden for de seneste 12 måneder
Andre aktuelle beviser for klinisk signifikant ukontrolleret sygdom/tilstande i ethvert legeme eller organsystem
Neurologisk eller psykiatrisk sygdom eller historie med stof- eller alkoholmisbrug hos en forsøgsperson eller dennes værge, som efter efterforskerens mening kunne forstyrre forsøgspersonens korrekte udfyldelse af protokolkravene
Deltagelse i en interventionsundersøgelse eller brugt et hvilket som helst forsøgslægemiddel til enhver sygdomstilstand inden for de sidste 30 dage

Potente cytokrom P450 3A4 (CYP3A4) hæmmere inden for de sidste 4 uger (f.eks. ritonavir, ketoconazol, itraconzol)

Tid til første hændelse i det sammensatte endepunkt af alvorlige astma-relaterede udfald i løbet af den 6-måneders undersøgelsesbehandlingsperiode blev analyseret ved hjælp af en Cox proportional hazards regressionsmodel. Et estimat af absolut risikoforskel og dets tilsvarende 95 % konfidensinterval (CI) blev også inkluderet.
