Detaillierte Beschreibung
Diese klinische Laborstudie wird doppelblind, placebokontrolliert sein und ein experimentelles Design innerhalb der Probanden verwenden.
Die Teilnehmer absolvieren 7 ambulante Sitzungen zur Medikamentenverabreichung, die aus der Selbstverabreichung von oralem Cannabis (0, 10 oder 25 mg THC) und Alkohol (entweder Placebo oder aktiv; BAC von 0,05 Prozent) bestehen; Die Teilnehmer erhalten immer sowohl ein alkoholisches Getränk (aktiv oder Placebo) als auch eine Dosis Cannabis (aktiv oder Placebo).
Die Teilnehmer werden auch eine Bedingung absolvieren, in der sie Alkohol (BAC: 0,08 Prozent) mit Placebo-Cannabis als Positivkontrolle verabreichen.
Zu den primären Ergebnissen gehören die Leistung bei Nüchternheitstests vor Ort, kognitive und psychomotorische Beeinträchtigungen, subjektive Arzneimittelwirkungen und simulierte Fahrleistung.
Blutkonzentrationen von THC und THC-Metaboliten werden ebenfalls bestimmt.

Einschlusskriterien:
- Eine schriftliche Einverständniserklärung abgegeben haben
- zwischen 21 und 50 Jahre alt sein
- Seien Sie in einem guten allgemeinen Gesundheitszustand, basierend auf einer körperlichen Untersuchung, Anamnese, Vitalfunktionen und Screening-Urin- und Bluttests
- Nicht schwanger sein oder stillen (falls weiblich). Alle Frauen müssen beim Screening-Besuch einen negativen Serum-Schwangerschaftstest und bei jedem Studienbesuch einen negativen Urin-Schwangerschaftstest haben.
- Haben Sie einen Body-Mass-Index (BMI) im Bereich von 19 bis 36 kg/m2
- Der Blutdruck beim Screening-Besuch überschreitet nicht einen systolischen Blutdruck (SBP) von 150 mmHg oder einen diastolischen Blutdruck (DBP) von 90 mmHg
- In den letzten 30 Tagen kein Blut gespendet haben.
- Melden Sie mindestens 2 Tage Rauschtrinken in den letzten 90 Tagen (mehr als 4 oder 5 Getränke bei einer einzigen Gelegenheit für Frauen bzw. Männer)
- Melden Sie ≥ 5 Cannabiskonsumenten im vergangenen Jahr
- Führen Sie beim Screening und vor Studiensitzungen einen negativen Urintest für den Konsum illegaler Drogen (ohne THC) und einen negativen Atemalkoholtest (0% BAC) durch
- Melden Sie mindestens 1 Fall von gleichzeitigem Alkohol- und Konsum im vergangenen Jahr.
Ausschlusskriterien:
- Konsum von psychoaktiven Drogen (außer Cannabis, Nikotin, Alkohol oder Koffein) im vergangenen Monat
- Aktuelle Verwendung von rezeptfreien (OTC) Arzneimitteln, Nahrungsergänzungsmitteln/Vitaminen oder verschreibungspflichtigen Medikamenten, die nach Ansicht des Prüfarztes oder des medizinischen Personals die Sicherheit des Teilnehmers beeinträchtigen
- Vorgeschichte oder aktuelle Hinweise auf einen signifikanten medizinischen Zustand
- Nachweis des aktuellen psychiatrischen Zustands [(MINI for Diagnostic and Statistical Manual (DSM)-V)]
- Erfüllen Sie die Kriterien für eine schwere Alkoholkonsumstörung (MINI für DSM-V)
- Clinical Institute Withdrawal Assessment for Alcohol scale (CIWA-Ar) score > 9
- Vorher in Behandlung wegen Alkohol- oder Cannabiskonsumstörung
- Konsum von Cannabis im Durchschnitt mehr als 2 Mal/Woche in den letzten 3 Monaten
- Leberfunktionstests mehr als 2x Normalbereich
- Teilnahme an einer anderen klinischen Studie oder Erhalt eines Arzneimittels als Teil der Forschung innerhalb der letzten 30 Tage
