Este estudo avaliará os efeitos individuais e interativos da cannabis oral e do álcool em medidas subjetivas e comportamentais de comprometimento.

Este estudo de laboratório clínico será duplo-cego, controlado por placebo e utilizará um projeto experimental intra-sujeitos.
Os participantes completarão 7 sessões de administração de medicamentos ambulatoriais que consistirão na autoadministração de cannabis oral (0, 10 ou 25 mg de THC) e álcool (placebo ou ativo; BAC de 0,05 por cento); os participantes sempre receberão uma bebida alcoólica (ativa ou placebo) e uma dose de cannabis (ativa ou placebo).
Os participantes também completarão uma condição na qual administrarão álcool (BAC: 0,08 por cento) com cannabis placebo, como controle positivo.
Os resultados primários incluem desempenho em testes de sobriedade de campo, comprometimento cognitivo e psicomotor, efeitos subjetivos de drogas e desempenho simulado ao dirigir.
As concentrações sanguíneas de THC e metabólitos de THC também serão determinadas.

Critério de inclusão:
- Forneceu consentimento informado por escrito
- Ter entre 21 e 50 anos
- Estar em boa saúde geral com base em um exame físico, histórico médico, sinais vitais e triagem de urina e exames de sangue
- Não estar grávida ou amamentando (se mulher). Todas as mulheres devem ter um teste de gravidez de soro negativo na visita de triagem e um teste de gravidez de urina negativo em cada visita do estudo.
- Ter um índice de massa corporal (IMC) na faixa de 19 a 36 kg/m2
- A pressão arterial na visita de triagem não excede a pressão arterial sistólica (PAS) de 150 mmHg ou a pressão arterial diastólica (DBP) de 90 mmHg
- Não ter doado sangue nos últimos 30 dias.
- Relatar pelo menos 2 dias de consumo excessivo de álcool nos últimos 90 dias (mais de 4 ou 5 doses em uma única ocasião para mulheres e homens, respectivamente)
- Relatar ≥ 5 usos de maconha no último ano
- Forneça teste de urina negativo para uso de drogas ilícitas (excluindo THC) e teste de álcool no ar expirado negativo (0% BAC) na triagem e antes das sessões de estudo
- Relate pelo menos 1 ocorrência simultânea de álcool e uso no último ano.
Critério de exclusão:
- Uso de drogas psicoativas (além de maconha, nicotina, álcool ou cafeína) no último mês
- Uso atual de medicamentos de venda livre (OTC), suplementos/vitaminas ou medicamentos prescritos que, na opinião do investigador ou da equipe médica, afetarão a segurança do participante
- Histórico ou evidência atual de condição médica significativa
- Evidência de condição psiquiátrica atual [(MINI for Diagnostic and Statistical Manual (DSM)-V)]
- Atende aos critérios para transtorno grave por uso de álcool (MINI para DSM-V)
- Avaliação da Abstinência do Instituto Clínico para Escala de Álcool (CIWA-Ar) pontuação > 9
- Esteve em tratamento anteriormente para transtorno por uso de álcool ou cannabis
- Uso de maconha, em média, mais de 2 vezes/semana nos últimos 3 meses
- Testes de função hepática mais de 2x a faixa normal
- Inscrição em outro ensaio clínico ou recebimento de qualquer medicamento como parte da pesquisa nos últimos 30 dias
- Pontuação de vocabulário Shipley <18 (corresponde ao nível de leitura da 5ª série).

Mean Peak Change From Baseline DRUID Application Global Impairment Score
Prazo: 7.5 hours, assessed at baseline (prior to drug administration) and again 1, 1.5, 2.5, 3.5, 4.5, 5.5, 6.5, and 7.5 hours after oral cannabis ingestion.

Acute cognitive and behavioral impairment will be assessed with global impairment score (range 0-100) on the DRUID app (higher scores indicate greater impairment).
Results are mean change from baseline.

Cumulative Score on Field Sobriety Tests

Impairment will be assessed using a battery of standard field sobriety tests including: the Horizontal Gaze Nystagmus Test (HGN), the Walk and Turn, the One Leg Stand, and the Modified Romberg Balance.
We will report the cumulative amount of clues observed across these tasks (out of a possible 0-22 clues).
Higher scores indicate greater/worse impairment.
These assessments were completed once after 7.5 hours.

There are 7 sedative-related questionnaire items, each presented on a scale of 0 (not at all) to 10 (extremely), which are integrated to produce an overall sedative score (0-70).
Higher scores indicating greater sedative effects of alcohol.
Scores presented show the change from baseline to 7.5 hours.

For the SHAS, participants are presented with 13 questionnaire items, displayed on a visual analog scale anchored from 0 (normal) to 10 (extremely), which assess subjective effects of alcohol.
These items are integrated to produce an overall SHAS score (0-130).
Higher score indicates greater effects of alcohol.

Driving Performance as Assessed by Standard Deviation of Lateral Position (SDLP)

The STISIM Drive® M4000-R Console system will be used to assess driving performance, a state-of-the-art technology that has been independently validated to reflect real-world driving conditions.
SDLP (measured in cm) was calculated across all drives to get a composite index of lateral control and reported as peak change from baseline.
SDLP is the gold standard of quantifying the magnitude of driving impairment from drugs and alcohol and has excellent predictive validity to actual driving.
Scores range from 0 to no upper limit.
Higher scores represent higher magnitude of driving impairment.

The composite drive score is derived by integrating various driving outcomes (see primary and secondary driving outcomes).
Higher z-score indicates worse performance.
The score is on a z-score scale, meaning a score of 1 equates to 1 standard deviation outside of the participants' mean baseline performance and a Z-score of 0 represents the mean baseline performance.
This was calculated across all drives and reported as peak change from baseline.
