Deze studie evalueert de individuele en interactieve effecten van orale cannabis en alcohol op subjectieve en gedragsmaten van beperkingen.

Deze klinische laboratoriumstudie zal dubbelblind, placebo-gecontroleerd zijn en zal een experimenteel ontwerp binnen de proefpersoon gebruiken.
Deelnemers zullen 7 poliklinische medicijntoedieningssessies voltooien die zullen bestaan uit zelftoediening van orale cannabis (0, 10 of 25 mg THC) en alcohol (hetzij placebo of actief; BAG van 0,05 procent); deelnemers krijgen altijd zowel een alcoholische drank (actief of placebo) als een dosis cannabis (actief of placebo).
Deelnemers voltooien ook een aandoening waarbij ze alcohol (BAC: 0,08 procent) toedienen met placebo-cannabis, als positieve controle.
Primaire uitkomsten zijn onder meer prestaties op nuchterheidstests in het veld, cognitieve en psychomotorische stoornissen, subjectieve drugseffecten en gesimuleerde rijprestaties.
Ook worden de bloedconcentraties van THC en THC-metabolieten bepaald.

Deelname Criteria
Onderzoekers zoeken naar mensen die aan een bepaalde beschrijving voldoen, de zogenaamde geschiktheidscriteria. Enkele voorbeelden van deze criteria zijn iemands algemene gezondheidstoestand of eerdere behandelingen.

Inclusiecriteria:
- Schriftelijke geïnformeerde toestemming hebben gegeven
- Tussen de 21 en 50 jaar oud zijn
- In goede algemene gezondheid verkeren op basis van een lichamelijk onderzoek, medische geschiedenis, vitale functies en screening van urine- en bloedtesten
- Niet zwanger zijn of borstvoeding geven (indien vrouw). Alle vrouwtjes moeten een negatieve serumzwangerschapstest hebben bij het screeningbezoek en een negatieve urinezwangerschapstest bij elk studiebezoek.
- Een body mass index (BMI) hebben in het bereik van 19 tot 36 kg/m2
- De bloeddruk bij het screeningsbezoek is niet hoger dan een systolische bloeddruk (SBP) van 150 mmHg of een diastolische bloeddruk (DBP) van 90 mmHg
- U heeft de afgelopen 30 dagen geen bloed gedoneerd.
- Rapporteer ten minste 2 dagen binge-drinken in de afgelopen 90 dagen (meer dan 4 of 5 drankjes bij een enkele gelegenheid voor respectievelijk vrouwen en mannen)
- Rapporteer ≥ 5 cannabisgebruiken in het afgelopen jaar
- Zorg voor een negatieve urinetest voor illegaal drugsgebruik (exclusief THC) en een negatieve ademalcoholtest (0% BAC) bij screening en vóór studiesessies
- Meld minimaal 1 keer gelijktijdig alcoholgebruik en gelijktijdig gebruik in het afgelopen jaar.
Uitsluitingscriteria:
- Psychoactief drugsgebruik (afgezien van cannabis, nicotine, alcohol of cafeïne) in de afgelopen maand
- Huidig gebruik van over-the-counter (OTC) medicijnen, supplementen / vitamines of voorgeschreven medicijnen die, naar de mening van de onderzoeker of medisch personeel, van invloed zijn op de veiligheid van de deelnemer
- Geschiedenis van of actueel bewijs van een significante medische aandoening
- Bewijs van huidige psychiatrische aandoening [(MINI for Diagnostic and Statistical Manual (DSM)-V)]
- Voldoen aan criteria voor ernstige alcoholgebruiksstoornis (MINI voor DSM-V)
- Clinical Institute Intrekkingsbeoordeling voor alcoholschaal (CIWA-Ar) score> 9
- Eerder in behandeling geweest voor een stoornis in het gebruik van alcohol of cannabis
- Gebruik van cannabis, gemiddeld meer dan 2 keer per week gedurende de afgelopen 3 maanden
- Leverfunctietests meer dan 2x normaal bereik
- Inschrijving in een ander klinisch onderzoek of ontvangen van een geneesmiddel als onderdeel van onderzoek in de afgelopen 30 dagen
- Shipley-woordenschatscore <18 (komt overeen met leesniveau 5e leerjaar).
