Este estudio de laboratorio clínico será doble ciego, controlado con placebo y utilizará un diseño experimental dentro de los sujetos.
Los participantes completarán 7 sesiones de administración de drogas para pacientes ambulatorios que consistirán en la autoadministración de cannabis oral (0, 10 o 25 mg de THC) y alcohol (ya sea placebo o activo; BAC de 0.05 por ciento); los participantes siempre recibirán tanto una bebida alcohólica (activo o placebo) como una dosis de cannabis (activo o placebo).
Los participantes también completarán una condición en la que administran alcohol (BAC: 0,08 por ciento) con cannabis placebo, como control positivo.
Los resultados primarios incluyen el desempeño en las pruebas de sobriedad en el campo, el deterioro cognitivo y psicomotor, los efectos subjetivos de las drogas y el desempeño de conducción simulada.
También se determinarán las concentraciones en sangre de THC y metabolitos de THC.

Criterios de inclusión:
- Haber dado su consentimiento informado por escrito
- Tener entre 21 y 50 años
- Gozar de buena salud general según un examen físico, historial médico, signos vitales y exámenes de detección de orina y sangre.
- No estar embarazada o amamantando (si es mujer). Todas las mujeres deben tener una prueba de embarazo en suero negativa en la visita de selección y una prueba de embarazo en orina negativa en cada visita del estudio.
- Tener un índice de masa corporal (IMC) en el rango de 19 a 36 kg/m2
- La presión arterial en la visita de selección no supera una presión arterial sistólica (PAS) de 150 mmHg o una presión arterial diastólica (PAD) de 90 mmHg
- No haber donado sangre en los últimos 30 días.
- Informar al menos 2 días de consumo excesivo de alcohol en los últimos 90 días (más de 4 o 5 tragos en una sola ocasión para mujeres y hombres, respectivamente)
- Informar ≥ 5 usos de cannabis en el último año
- Proporcionar prueba de orina negativa para uso de drogas ilícitas (excluyendo THC) y prueba de alcohol en aliento negativa (0% BAC) en la selección y antes de las sesiones de estudio
- Informe al menos 1 instancia de alcohol y consumo simultáneos en el último año.
Criterio de exclusión:
- Consumo de drogas psicoactivas (aparte de cannabis, nicotina, alcohol o cafeína) en el último mes
- Uso actual de medicamentos de venta libre (OTC), suplementos/vitaminas o medicamentos recetados que, en opinión del investigador o del personal médico, afectarán la seguridad del participante
- Historia o evidencia actual de una condición médica importante
- Evidencia de condición psiquiátrica actual [(MINI para el Manual de Diagnóstico y Estadística (DSM)-V)]
- Cumplir con los criterios para el trastorno grave por consumo de alcohol (MINI para DSM-V)
- Puntaje de la escala Clinical Institute Withdrawal Assessment for Alcohol (CIWA-Ar) > 9
- Ha estado en tratamiento anteriormente por trastorno por consumo de alcohol o cannabis
- Uso de cannabis, en promedio, más de 2 veces por semana durante los últimos 3 meses
- Pruebas de función hepática más de 2 veces el rango normal

Higher scores indicate greater/worse impairment.

SDLP is the gold standard of quantifying the magnitude of driving impairment from drugs and alcohol and has excellent predictive validity to actual driving.
