Un estudio de fase 1b/2 de dosis única y múltiple para evaluar la seguridad, la tolerabilidad, la actividad clínica, la farmacocinética, la farmacodinámica y la inmunogenicidad de STAR-0215 en participantes con angioedema hereditario (ensayo ALPHA-STAR)
El objetivo de este ensayo clínico es probar el fármaco STAR-0215 en participantes con angioedema hereditario (AEH).
Un grupo de participantes recibirá 1 dosis de STAR-0215 y el otro grupo recibirá 2 dosis de STAR-0215.
Los investigadores estudiarán los efectos de STAR-0215 en participantes con AEH, ya que es la primera vez que se administra el medicamento a participantes con AEH.

Descripción detallada
Este es un ensayo de dosis única y múltiple de fase 1b/2 que evalúa la seguridad, la tolerabilidad, la actividad clínica, la farmacocinética, la farmacodinámica y la inmunogenicidad de la administración subcutánea de STAR-0215 en participantes con AEH tipo I o tipo II en cohortes de 2 dosis.
La primera cohorte recibirá 1 dosis de STAR-0215; la segunda cohorte recibirá 2 dosis secuenciales.
Este es el primer ensayo de STAR-0215 en participantes con AEH y la primera evaluación de un régimen de dosis múltiples.

Criterios de inclusión:
Diagnóstico documentado de AEH (tipo I o II). Se debe cumplir con lo siguiente:
una. Antecedentes clínicos documentados consistentes con AEH (por ejemplo, episodios de inflamación subcutánea o mucosa no pruriginosa sin urticaria acompañante).
- Experimentó al menos 4 ataques de AEH en los 12 meses anteriores a la selección, por informe de participante.
- Experimentó al menos 2 ataques de AEH durante el período inicial, según lo confirmó un investigador en función del cumplimiento de la definición de ataque de AEH especificada en el protocolo.
Criterio de exclusión:
- Cualquier diagnóstico concomitante de otra forma de angioedema crónico, como deficiencia adquirida del inhibidor de C1, AEH con C1-INH normal (también conocido como AEH tipo III), angioedema idiopático o angioedema asociado con urticaria.
Uso de terapias prescritas para la prevención de ataques de AEH antes de la Selección:
- lanadelumab dentro de los 90 días
- berotralstat dentro de los 21 días
- todas las demás terapias profilácticas, dentro de los 7 días
- Cualquier exposición a inhibidores de la enzima convertidora de angiotensina o cualquier medicamento que contenga estrógeno con absorción sistémica (como anticonceptivos hormonales o terapia de reemplazo hormonal) dentro de los 28 días anteriores a la Selección.
- Cualquier exposición a andrógenos (por ejemplo, estanozolol, danazol, oxandrolona, metiltestosterona, testosterona) dentro de los 7 días anteriores a la Selección.

An adverse event was any untoward medical occurrence in a clinical investigation participant who was administered a pharmaceutical product that did not necessarily have a causal relationship with the treatment.
A treatment-emergent adverse event was defined as any adverse event with an onset at the time of or following the start of treatment with study drug, or medical conditions present before the start of treatment that increased in severity or relationship at the time of or following the start of treatment.
