Dit is een fase 2, multicenter, gerandomiseerde, parallelle, 3-armige, placebogecontroleerde studie om de werkzaamheid en veiligheid van CDR132L te beoordelen bij patiënten met een verminderde linkerventrikelejectiefractie (LVEF) (≤ 45%) na een myocardinfarct (MI). Dit onderzoek bestaat uit een screeningperiode (die minimaal 3 dagen na de diagnose MI moet plaatsvinden), een dubbelblinde periode van 6 maanden en een verlengingsperiode van 6 maanden met het einde van het onderzoek (EOS) op dag 360/maand 12 .

Twee doseringen van CDR132L zullen worden getoetst aan placebo op hun effecten op patiënten die net een hartaanval hebben gehad naast de standaardzorg. Het doel van de studie is om aan te tonen dat CDR132L veilig en effectief is om hartfalen bij dergelijke patiënten te verbeteren.

Belangrijkste opnamecriteria:

  1. Mannelijke of vrouwelijke patiënten van ≥ 30 tot ≤ 80 jaar op de datum van ondertekening van de geïnformeerde toestemming, gedefinieerd als het begin van de screeningperiode.
  2. Spontaan acuut mycardiaal infarct (AMI) (type I) gebaseerd op de universele MI-definitie met randomisatie die niet later dan 14 dagen na de diagnose van het indexgebeurtenis optreedt.
  3. Patiënt met een LVEF ≤ 45% zoals gemeten door ECHO na MI-diagnose (STEMI of NSTEMI).
  4. Patiënt met eerdere MI-gebeurtenissen in de geschiedenis kan worden opgenomen.
  5. Patiënt met een lichaamsgewicht van ≤ 120 kg.
  6. Niveau N-terminaal pro-B-type natriuretisch peptide ≥ 125 pg/ml en < 8000 pg/ml bij screening.
  7. Patiënt met STEMI/NSTEMI die voor dit evenement percutane coronaire interventie onderging.

Uitsluitingscriteria:

  1. Een vrouw in de vruchtbare leeftijd (WOCBP).
  2. Patiënt met HF van niet-ischemische oorsprong; bijv. myocarditis, alcoholische cardiomyopathie.
  3. Patiënt met New York Heart Association (NYHA) klasse IV bij screening of randomisatie.
  4. De patiënt heeft een geplande hartinterventie (angiogram zonder angioplastiek is acceptabel) of een andere geplande operatie na de screeningperiode.
  5. Patiënt heeft een ernstige hartklepaandoening.
  6. Patiënt heeft systolische bloeddruk < 90 mmHg of > 180 mmHg, diastolische bloeddruk < 50 mmHg of > 110 mmHg, en/of hartslag < 50 of > 100 slagen/minuut bij screening of randomisatie.
  7. Patiënt met een geschatte glomerulaire filtratiesnelheid < 30 ml/min/1,73 m2 of bij dialyse.
  8. Patiënt met leverinsufficiëntie geclassificeerd als Child-Pugh B of C.
  9. Patiënt heeft een medische voorgeschiedenis van ziekte(n) die de bloed-hersenbarrière aantasten, bijv. beroerte binnen 6 maanden of multiple sclerose.
  10. Patiënt heeft een medische voorgeschiedenis van bloedingsstoornissen of heeft trombocytopenie (bloedplaatjes < 100.000/μL).
  11. Patiënt heeft slecht gecontroleerde diabetes zoals vastgesteld door de onderzoeker.
  12. Patiënt heeft een voorgeschiedenis of aanwezigheid van een van de volgende hartaandoeningen: bekende structurele hartafwijkingen buiten HF, familiegeschiedenis van lang-QT-syndroom, cardiale syncope of recidiverende idiopathische syncope.
