Aktualne dane wskazują, że preparaty z dziurawca mogą indukować wątrobowe enzymy cytochromu P450 i białka transportowe. Może to powodować interakcje z lekami.
Projekt badania jest standardem dla badań DDI i opiera się na wytycznych regulacyjnych Agencji ds. Żywności i Leków (FDA) oraz Europejskiej Agencji Leków (EMA).
Podejście koktajlowe obejmujące podawanie wielu leków-sond specyficznych dla cytochromu P450 (CYP) lub P-glikoproteiny (P-gp) jest stosowane do jednoczesnej oceny aktywności tych enzymów i transportera P-gp.

Kryteria przyjęcia:
- pisemna świadoma zgoda
- Mężczyźni lub kobiety rasy kaukaskiej w wieku od ≥18 do ≤55 lat
- Fizycznie i psychicznie zdrowy
- BMI między ≥19 a ≤29 kg/m2 i masa ciała ≤90 kg
- Niepalący
- W przypadku kobiet test ciążowy podczas badania przesiewowego i przy przyjęciu musi być ujemny
Kryteria wyłączenia:
- Znana lub podejrzewana nadwrażliwość na badane leki
- historia, wszelkie istotne klinicznie choroby
- Pozytywny wynik testu na wirusowe zapalenie wątroby typu B, wirusowe zapalenie wątroby typu C lub badanie przesiewowe w kierunku HIV
- Znana fotonadwrażliwość

The "number" as provided in the tables is the ratio (in %) of geometric LS means of AUC of day 17 dosing samples (probe drug cocktail plus Ze117) vs. Day 1 dosing samples (probe drug cocktail alone). A one-way ANOVA model (with terms for subject and treatment) was applied to analyze the effect of Ze117 on the respective probe drug. Log transformed (base e) parameters were used within the model. The estimated treatment difference between day 17 and day 1 was transformed back to original scale and presented as the ratio between treatments including the corresponding 90% confidence interval. Effect limits were predefined as follows: (1) Mild effect limits (50-200%), extended bioequivalence limits (70-143%), bioequivalence limits (80-125%). |
