Description détaillée
Les données actuelles indiquent que les préparations de millepertuis peuvent induire des enzymes hépatiques du cytochrome P450 et des protéines de transport. Cela peut entraîner des interactions médicamenteuses.
La conception de l'étude est standard pour les études DDI et est basée sur les directives réglementaires de la Food and Drug Administration (FDA) et de l'Agence européenne des médicaments (EMA).
Une approche cocktail impliquant l'administration de plusieurs médicaments sondes spécifiques au cytochrome P450 (CYP) ou à la glycoprotéine P (P-gp) est utilisée pour évaluer simultanément les activités de ces enzymes et du transporteur P-gp.

Critère d'intégration:
- consentement éclairé écrit
- Sujets caucasiens de sexe masculin ou féminin âgés de ≥ 18 à ≤ 55 ans
- Physiquement et mentalement sain
- IMC entre ≥19 et ≤29 kg/m2 et poids corporel ≤90 kg
- Non fumeur
- Si femme, le test de grossesse au dépistage et à l'admission doit être négatif
Critère d'exclusion:
- Hypersensibilité connue ou suspectée aux médicaments à l'étude
- antécédents de toute maladie cliniquement significative
- Test positif de l'hépatite B, de l'hépatite C ou du dépistage du VIH
- Photohypersensibilité connue

The "number" as provided in the tables is the ratio (in %) of geometric LS means of AUC of day 17 dosing samples (probe drug cocktail plus Ze117) vs. Day 1 dosing samples (probe drug cocktail alone). A one-way ANOVA model (with terms for subject and treatment) was applied to analyze the effect of Ze117 on the respective probe drug. Log transformed (base e) parameters were used within the model. The estimated treatment difference between day 17 and day 1 was transformed back to original scale and presented as the ratio between treatments including the corresponding 90% confidence interval. Effect limits were predefined as follows: (1) Mild effect limits (50-200%), extended bioequivalence limits (70-143%), bioequivalence limits (80-125%). |
