一项针对患有 ACS 的受试者的双盲随机平行组安慰剂对照研究。 除了阿托伐他汀 QD 和标准护理外，多达 6500 名受试者将被随机分配接受 A-002 500 mg 每日一次 (QD) 或安慰剂片剂。 治疗将持续 16 周。 如果受试者的 LDL-C ≥ 100 mg/dL，则应在 8 周后调整阿托伐他汀的剂量，否则必须在整个 16 周的研究期间保持稳定。 所有登记受试者的生存状态将在他们完成研究后 6 个月确定。

  1. ≥40岁的男性和女性
  2. 受试者的书面知情同意书

  3. 不稳定型心绞痛、非 ST 段抬高型心肌梗死 (NSTEMI) 或 ST 段抬高型心肌梗死 (STEMI) 的诊断

    不稳定型心绞痛定义为：

    • 胸痛症状为缺血或心绞痛，在休息或最小运动时发生，频率或严重程度增加，持续时间 > 10 分钟，并与住院前 24 小时内的心肌缺血一致，并且
    • 至少 2 个相邻导联出现新的或动态的 ST 段压低或明显的 T 波倒置改变，并且
    • 此外，符合上述不稳定型心绞痛标准的受试者还必须具有肌钙蛋白 I、肌钙蛋白 T 或 CKMB 高于 LLD 但低于参考上限 (URL) 的第 99 个百分位数，并且不是由于心脏复律或潜在的心血管疾病（CHF、心肌病）或肾病

    NSTEMI 定义为：

  2. 在过去 5 年内接受过除皮肤基底细胞癌或宫颈原位癌以外的其他癌症治疗的受试者，并采取了除轻微、完全手术切除或放射治疗以外的措施（例如 化疗）
  3. 存在任何伴有肝硬化的严重肝病、活动性肝炎、活动性慢性肝炎、谷丙转氨酶 (ALT) 或天冬氨酸转氨酶 (AST) >3 x ULN、胆道梗阻伴高胆红素血症（总胆红素 >2 x ULN）
  4. 活动性胆囊炎、胆囊症状或任何肝胆异常
  5. 存在严重肾功能损害（肌酐清除率 [CrCl] <30 mL/min 或肌酐 >3 x ULN）、肾病综合征或接受透析的受试者
  6. 不受控制的糖尿病（已知血红蛋白 A1c [HbA1c] 在筛选前的最后 1 个月内 >11%）
  7. 在研究期间正在哺乳、怀孕或打算怀孕的女性，或在筛选评估期间妊娠试验呈阳性的具有生育潜力的女性。 有生育能力的女性还必须在研究期间和治疗完成后 1 个月内使用可靠的节育方法。 本研究的可靠方法定义为以下之一：口服或注射避孕药、宫内节育器 (IUD)、避孕植入物、输卵管结扎术、子宫切除术、双屏障方法（带有杀精泡沫或果冻的隔膜，或避孕套）。
  8. 在研究开始后 1 年内有酒精或药物滥用史的受试者
  9. 居住在离参与中心太远或无法返回进行后续访问的受试者
