涉及晚期实体瘤患者的 I 期研究数据表明,LBH589 的每周 MWF 时间表在药效学和临床上均具有活性。 在 20 mg 和 30 mg 剂量下,50% 的患者在外周血细胞中检测到组蛋白乙酰化长达 72 小时(MWF 每周计划给药之间的最长持续时间),并且在 2 名和 3 名患者中观察到 CR 和 PR分别与皮肤T细胞淋巴瘤有关。 3 级疲劳是 2 名接受 40 mg 水平治疗的患者和 1 名接受 60 mg 水平治疗的患者的剂量限制性毒性 (DLT)。 40 mg 水平似乎是可以耐受的,并且被认为最有可能在实体瘤中具有活性。

纳入标准
- 年龄 ≥ 18 岁。
- 经组织学证实的晚期转移性或不可切除的软组织肉瘤,不包括骨肉瘤。
- 无论是在辅助治疗中还是在转移性/晚期疾病中,既往使用含多柔比星的方案进行过治疗。 如果给予多柔比星作为辅助治疗,如果在辅助治疗后一年内复发,则患者可能会被纳入。 如果在辅助治疗完成后一年以上发生复发,则患者必须接受过一种先前的转移性疾病治疗方案。 患者可能已经接受过一种或多种先前的治疗。 患有性索肿瘤的患者可以在先前用含铂方案治疗后包括在内(该患者亚组不需要用含阿霉素的方案进行预处理)。
- 患者至少有一处可测量的淋巴结病变部位在基线时最长横径 ≥ 2.0 cm,并且通过 CT 扫描确定在至少两个垂直维度上可清楚地测量(仅当无法进行 CT 扫描时才允许使用 MRI)。
- ECOG 体能状态 (PS) ≤ 2。
