这是一项 3 期、开放标签、随机临床试验，评估 gedatolisib 加氟维司群联合 CDK4/6 抑制剂（研究者选择 palbociclib 或 ribociclib）治疗晚期（无法手术）或转移性激素患者的有效性和安全性受体阳性、人表皮生长因子受体 2 阴性 (HR+/HER2-) 乳腺癌，其疾病在辅助治疗期间或 12 个月内出现进展接受内分泌治疗（AI 或他莫昔芬），并且之前未接受过 ABC 全身治疗。
Gedatolisib 是一种静脉注射的泛 PI3K/mTOR 抑制剂。
Palbociclib 是一种 CDK4/6 抑制剂。
氟维司群是一种选择性雌激素受体降解剂（SERD）。
在完成安全磨合阶段以确定 ribociclib 的 gedatolisib 剂量后，受试者将根据其 PIK3CA 状态被分配到 2 个队列中的一组，然后随机分配到研究治疗组（gedatolisib 联合氟维司群和 ribociclib 或 palbociclib）或标准护理对照组（氟维司群和 ribociclib 或 palbociclib）。

- 经组织学或细胞学确诊的转移性或局部晚期 HR+/HER2- 乳腺癌
- 成年女性、绝经前和/或绝经后以及成年男性。 如果愿意接受 LHRH 激动剂治疗的绝经前（和围绝经期）女性可以参加。 患者应在第 1 周期第 1 天之前或之时开始 LHRH 激动剂的伴随治疗，并且必须愿意在研究期间继续进行。
- 对有生育能力的女性进行阴性妊娠试验。 未进行绝育手术的女性受试者必须从筛查到最后一次研究治疗后 2 年内使用医学上有效的避孕方法。
- 在完成（新）辅助 ET 期间或完成后 12 个月内疾病进展。

允许的先前治疗：
