Dette er et multicenter, to-trins, randomiseret, kontrolleret, åbent fase 3-studie, der sammenligner effektiviteten og sikkerheden af ​​iberdomid i kombination med dexamethason og daratumumab (IberDd) versus daratumumab, bortezomib og dexamethason (DVd) hos deltagere med tilbagefald. eller refraktær myelomatose (RRMM). Ca. 200 patienter randomiseret i trin 1 til et af tre iberdomid-dosisniveauer på 1, 1,3 eller 1,6 mg i kombination med daratumumab og dexamethason (behandlingsarme A1, A2 eller A3) eller til DVd-komparatorarmen (behandlingsarm B) .

I fase 2 af studiet vil cirka 664 yderligere forsøgspersoner blive randomiseret 1:1 mellem 2 behandlingsarme:

  • Cirka 332 forsøgspersoner vil blive randomiseret til at modtage behandlingsarm A (IberDd)
  • Cirka 332 forsøgspersoner vil blive randomiseret til at modtage behandlingsarm B (DVd)

Deltagere i begge behandlingsarme vil fortsætte med at modtage behandling indtil bekræftet progressiv sygdom (PD), uacceptabel toksicitet eller tilbagetrækning af samtykke. Inklusionskriterier:

  • Dokumenteret diagnose af myelomatose (MM) og målbar sygdom
  • Modtaget 1 til 2 tidligere linjer med anti-myelomterapi
  • Skal have dokumenteret sygdomsprogression under eller efter deres sidste anti-myelombehandling
  • Eastern Cooperative Oncology Group (ECOG) præstationsstatusscore på 0, 1 eller 2

Ekskluderingskriterier:

  • Enhver tilstand, der forstyrrer evnen til at fortolke data fra undersøgelsen
  • Har plasmacelleleukæmi, Waldenstroms makroglobulinæmi eller POEMS-syndrom (polyneuropati, organomegali, endokrinopati, monoklonalt protein og hudforandringer) eller klinisk signifikant amyloidose
  • Kendt involvering af centralnervesystemet med MM
