Dette er en multisenter, to-trinns, randomisert, kontrollert, åpen fase 3-studie som sammenligner effekten og sikkerheten til iberdomid i kombinasjon med deksametason og daratumumab (IberDd) versus daratumumab, bortezomib og deksametason (DVd) hos deltakere med tilbakefall. eller refraktært multippelt myelom (RRMM). Omtrent 200 pasienter randomisert i stadium 1 til ett av tre iberdomiddosenivåer på 1, 1,3 eller 1,6 mg i kombinasjon med daratumumab og deksametason (behandlingsarmer A1, A2 eller A3), eller til DVd-komparatorarmen (behandlingsarm B) .

I trinn 2 av studien vil omtrent 664 tilleggspersoner bli randomisert 1:1 mellom 2 behandlingsarmer:

  • Omtrent 332 forsøkspersoner vil bli randomisert til å motta behandlingsarm A (IberDd)
  • Omtrent 332 forsøkspersoner vil bli randomisert til å motta behandlingsarm B (DVd)

Deltakere i begge behandlingsarmene vil fortsette å motta behandling inntil bekreftet progressiv sykdom (PD), uakseptabel toksisitet eller tilbaketrekking av samtykke. For å sikre nøyaktighet og fullstendighet av den primære endepunktsvurderingen av progresjonsfri overlevelse (PFS), vil deltakere som permanent avbryter studiebehandlingen av en eller annen grunn, annet enn bekreftet PD eller tilbaketrekking av samtykke, fortsatt følges for sykdomsvurdering.

Inklusjonskriterier:

  • Dokumentert diagnose av myelomatose (MM) og målbar sykdom
  • Mottatt 1 til 2 tidligere linjer med myelombehandling
  • Må ha dokumentert sykdomsprogresjon under eller etter siste antimyelomregime
  • Eastern Cooperative Oncology Group (ECOG) resultatstatusscore på 0, 1 eller 2

Ekskluderingskriterier:

  • Enhver tilstand som forstyrrer evnen til å tolke data fra studien
  • Har plasmacelleleukemi, Waldenstroms makroglobulinemi eller POEMS-syndrom (polynevropati, organomegali, endokrinopati, monoklonalt protein og hudforandringer), eller klinisk signifikant amyloidose
  • Kjent sentralnervesystempåvirkning med MM
  • Tidligere behandling med iberdomid
