Une étude pour évaluer l'innocuité et l'efficacité de l'IPN10200 chez les participants adultes présentant des rides faciales supérieures modérées à sévères (LANTIC)
Une étude de phase Ib/II, multicentrique, en double aveugle, randomisée, contrôlée par placebo, d'escalade de dose et de recherche de dose pour évaluer l'innocuité et l'efficacité de l'IPN10200 dans l'amélioration de l'apparence des rides supérieures du visage modérées à sévères chez les adultes
Le but de cette étude est d'évaluer le profil d'innocuité et d'efficacité de doses croissantes d'IPN10200 par rapport au placebo, dans le but de découvrir la ou les doses qui offrent le meilleur profil d'efficacité/innocuité lorsqu'elles sont utilisées pour le traitement d'une forme modérée à sévère. Lignes supérieures du visage.
Cette étude se déroulera en trois étapes. L'étude complète (y compris toutes les étapes) comptera un maximum de 448 participants. Le protocole est actuellement approuvé jusqu'à l'étape 1, l'étape 1 et l'étape 1/étape 2.
Stade 1 (phase Ib & II)
- Étape 1 (Phase Ib) : une première étape d'escalade de dose chez l'homme chez les participants atteints de rides glabellaires (GL) modérées à sévères
- Étape 2 (Phase II) : étape de détermination de la dose chez les participants atteints de GL modérée à sévère par rapport à Dysport
Étape 2 (phase II)
- Une évaluation de l'efficacité et de l'innocuité de l'IPN10200 dans l'une des régions suivantes : GL + lignes frontales (FHL) ou lignes canthales latérales (LCL)
Étape 3 (phase II)
- Une évaluation de la sécurité et de l'efficacité de l'IPN10200 dans les trois régions (GL, FHL et LCL)

Critère d'intégration:
- Le participant doit être âgé de 18 à 65 ans inclus au moment de la signature du consentement éclairé.
- GL modérée ou sévère (grade 2 ou 3) à la contraction maximale au départ, telle qu'évaluée par l'ILA à l'aide d'une échelle photographique validée à 4 points.
- GL modérée ou sévère (grade 2 ou 3) à la contraction maximale au départ, telle qu'évaluée par le SSA à l'aide d'une échelle catégorielle validée à 4 points.
- Insatisfaits ou très insatisfaits (2e ou 3e année) de leurs lignes de base, telles qu'évaluées par le SLS.
Critère d'exclusion:
- Une infection active ou d'autres problèmes de peau dans la partie supérieure du visage, y compris la zone GL, FHL et LCL (par ex. lésions acnéiques aiguës ou ulcères).
- Antécédents de blépharoplastie des paupières ou de lifting des sourcils au cours des 5 dernières années
- Une histoire de paralysie du nerf facial.
- Asymétrie faciale marquée, ptose, dermatochalase excessive, cicatrices cutanées profondes ou peau sébacée épaisse.
- Toute condition médicale connue qui peut exposer le participant à un risque accru en ce qui concerne l'exposition à la BoNT de tout sérotype (c'est-à-dire myasthénie grave, syndrome d'Eaton-Lambert, sclérose latérale amyotrophique, etc.)
- A la maladie COVID-19 ou un test SARS-CoV-2 positif, ou la présence de toute autre condition (par ex. trouble neuromusculaire ou autre trouble pouvant interférer avec la fonction neuromusculaire)
- Traitement antérieur avec n'importe quel sérotype BoNT pour le stade 1 / étape 1 ou tout traitement récent (au cours des 6 derniers mois avant la ligne de base) avec n'importe quel sérotype BoNT pour le stade 1 / étape 2.
- Tout traitement préalable avec des charges permanentes dans le haut du visage, y compris la zone GL, FHL et LCL.
- Tout traitement préalable avec des produits de comblement dermique longue durée dans le haut du visage y compris la zone GL au cours des 3 dernières années et/ou abrasions cutanées/resurfacing (quelle que soit la technique interventionnelle utilisée) au cours des 5 dernières années, ou photorajeunissement ou intervention laser cutanée/vasculaire dans les 12 mois précédant la référence.
- Toute chirurgie esthétique faciale prévue pendant l'étude.
- Utilisation d'un traitement concomitant qui, de l'avis de l'investigateur, interférerait avec l'évaluation de l'innocuité ou de l'efficacité de l'intervention à l'étude, y compris les médicaments affectant les troubles de la coagulation (par ex. agents antiplaquettaires et/ou anticoagulants administrés pour le traitement ou la prévention des maladies cardiovasculaires/cérébrovasculaires).
- Utilisation de médicaments qui affectent la transmission neuromusculaire, tels que les agents non dépolarisants de type curare, les lincosamides, les polymyxines, les anticholinestérases et les antibiotiques aminoglycosides, au cours des 30 derniers jours précédant la ligne de base.
- Utilisation de tout dispositif expérimental dans les 30 jours ou utilisation de tout traitement avec un médicament expérimental dans les cinq fois la demi-vie terminale documentée du médicament respectif ou de ses métabolites ou si la demi-vie est inconnue dans les 30 jours précédant le début de l'étude ( avant l'étude de référence) et pendant la conduite de l'étude.
- Connu positif pour l'antigène de l'hépatite B, ou l'anticorps du virus de l'hépatite C, ou pour le virus de l'immunodéficience humaine (VIH) ou un diagnostic de syndrome d'immunodéficience acquise.
- Trouble anxieux important diagnostiqué cliniquement ou tout autre trouble psychiatrique important (p. dépression) qui pourrait interférer avec la participation du participant à l'étude
- Une incapacité à réduire considérablement les GL et/ou les rides horizontales du front, même en les écartant physiquement, comme déterminé par l'investigateur.
- Allergie ou hypersensibilité connue à la BoNT ou à l'un des excipients de l'IPN10200 ou du Dusport/Azzalure, ou allergie aux protéines de lait de vache.
- Antécédents d'abus de drogue ou d'alcool
- Les femmes enceintes, les femmes qui allaitent, les femmes préménopausées ou les femmes en âge de procréer qui ne souhaitent pas pratiquer une méthode de contraception très efficace
- Participants masculins qui ne sont pas vasectomisés et qui ont des partenaires féminines en âge de procréer et qui ne sont pas disposés à utiliser des préservatifs avec spermicide tout au long de leur participation à l'étude.
