Questo è uno studio clinico di Fase 3, multicentrico, randomizzato, in doppio cieco, a gruppi paralleli e controllato con placebo, volto a valutare l'efficacia e la sicurezza di HBS-301 nel trattamento dei sintomi dell'ipersonnia idiopatica (IH), inclusi l'eccessiva sonnolenza diurna (EDS), l'inerzia del sonno e la stanchezza, in partecipanti adulti (età ≥18 anni) con IH.

Si prevede di randomizzare circa 248 partecipanti nello studio. Lo studio consisterà in un Periodo di Screening/Linea di Base (fino a 28 giorni), un Periodo di Trattamento in Doppio Cieco (8 settimane), un Periodo facoltativo di Estensione in Aperto (OLE) (1 anno) e 30 giorni di follow-up per la sicurezza.

Descrizione

Criteri di inclusione:

  • Presenta una diagnosi documentata attuale di IH secondo i criteri della Classificazione Internazionale dei Disturbi del Sonno, Terza Edizione (ICSD-3) o della Revisione Testuale (ICSD-3-TR) con polisonnogramma (PSG) confermativo con test di latenza multipla del sonno (MSLT); e, se applicabile, un rapporto di actigrafia con registro del sonno in archivio che ha portato alla diagnosi ed è stato completato negli ultimi 10 anni.
  • Presenta sintomi di IH da moderati a molto gravi.
  • Se assume un farmaco concomitante cronico permesso o un integratore, inclusi antidepressivi non vietati, deve essere a un dosaggio stabile da almeno 3 mesi prima dello Screening e accettare di continuare a quel dosaggio stabile per il Periodo di Trattamento in Doppio Cieco dello studio. Qualsiasi trattamento che potrebbe influenzare la sonnolenza diurna (inclusi ma non limitati a stimolanti, modafinil e armodafinil) usato su base al bisogno non è permesso.
  • Per i partecipanti in trattamento per l'apnea ostruttiva del sonno (OSA) o altre condizioni di ipoventilazione, devono essere conformi al loro dispositivo medico o dispositivo orale come determinato dallo Sperimentatore. I partecipanti devono mantenere la conformità al trattamento OSA per tutta la durata dello studio.

Criteri di esclusione:

  • Presenta ipersonnia dovuta a un altro disturbo medico.
  • Ha una storia di uso di pitolisant entro 5 emivite prima dello Screening.
  • Presenta una diagnosi primaria di malattia psichiatrica non ben controllata.
  • Ha una storia di compromissione epatica moderata o grave.
  • Presenta una velocità di filtrazione glomerulare stimata (eGFR) corretta per la superficie corporea (BSA) <60 mL/min.
  • Ha una storia nota di sindrome del QT lungo o qualsiasi storia significativa di una grave anomalia dell'elettrocardiogramma (ECG).
