Prospektywne badanie obserwacyjne młodzieży z ciężkim atopowym zapaleniem skóry pomimo mniej rozległych zmian skórnych (wskaźnik obszaru wyprysku i ciężkości < 16) otrzymujących dupilumab
Badania kliniczne wykazały znaczną poprawę objawów podmiotowych i przedmiotowych atopowego zapalenia skóry (AZS) u młodzieży leczonej dupilumabem, przy dobrym profilu bezpieczeństwa. Do tych badań klinicznych włączono wyłącznie pacjentów z wskaźnikiem obszaru i ciężkości wyprysku (EASI) większym lub równym (≥) 16, a skuteczność w obszarach wrażliwych/widocznych nie była szczegółowo oceniana. Aby lepiej zrozumieć potencjalne korzyści kliniczne dupilumabu w tych populacjach, konieczne są zatem dalsze dane dotyczące skuteczności dupilumabu u młodzieży z umiarkowanym do łagodnego wynikiem EASI i silnym świądem i/lub miejscową chorobą Alzheimera.
Jest to włoskie wieloośrodkowe, 52-tygodniowe badanie obserwacyjne (nieinterwencyjne), w ramach którego zgromadzone zostaną dane dotyczące charakterystyki nastolatków (w wieku od 12 do 17 lat) cierpiących na ciężką chorobę Alzheimera z wynikiem EASI mniejszym niż (<) 16, kwalifikujących się do systemowe leczenie dupilumabem zgodnie z włoskimi kryteriami refundacyjnymi. W ramach badania zbadana zostanie rzeczywista skuteczność i bezpieczeństwo dupilumabu w tej populacji, wpływ dupilumabu na swędzenie (świąd), sen, jakość życia i powiązane wyniki, zlokalizowane AZS w obszarach wrażliwych/widocznych oraz na współistniejące schorzenia atopowe u młodzieży. uczestników otrzymujących dupilumab z powodu AD. Dokumentacja udokumentuje również satysfakcję z leczenia dupilumabem i przerwanie leczenia dupilumabem u uczestników badania.

Kryteria przyjęcia:
- Mężczyzna lub kobieta, w wieku od 12 do 17 lat w momencie wizyty początkowej
- Pacjenci z chorobą Alzheimera, którym przepisano dupilumab zgodnie z kryteriami refundacyjnymi Agenzia Italiana del Farmaco (AIFA) i spełniający następujące kryteria:
Pacjenci z EASI<16 i
- Wskaźnik jakości życia w dermatologii dziecięcej (CDLQI) ≥ 10 lub
- Numeryczna skala oceny szczytowego świądu (PP-NRS) ≥ 7 lub
lokalizacja w obszarach widocznych lub wrażliwych (głowa/szyja/dłonie lub narządy płciowe)
- Pacjenci są w stanie zrozumieć i wypełnić kwestionariusze związane z badaniem
- Pod warunkiem podpisanej świadomej zgody lub zgody rodziców/prawnie akceptowanego przedstawiciela i zgody pacjenta, jeśli ma to zastosowanie
Kryteria wyłączenia:
- Wcześniejsze stosowanie dupilumabu w ciągu 6 miesięcy przed przystąpieniem do badania
- Pacjenci biorący obecnie udział w jakimkolwiek interwencyjnym badaniu klinicznym modyfikującym opiekę nad pacjentem
- Każdy stan, który w opinii Badacza może zakłócać zdolność pacjenta do udziału w badaniu (np. nadużywanie substancji psychoaktywnych)

The PGA score is a measure used in clinical practice and clinical trials to assess the severity of a particular disease.
The PGA chosen for this study ranges from 0-5 point, and the following categories will be used: 0 = clear, 1 = almost clear, 2 = mild, 3 = moderate, 4 = severe, 5 = very severe.
In this study, the Investigator will be evaluating four PGA scores, including PGA face, PGA feet, PGA hands, and PGA genitalia, in order to assess the severity of the localized lesions in AD patients at baseline and at week 4, 16 and 52.

The PAQ is a 6-item measure for parents or caregivers of children aged 0-11 years with clinician-diagnosed asthma, or those with recurrent viral-associated wheezing with risk factors for persistent asthma, to assess both symptom control and asthma-specific HRQoL impact.
The PAQ features two items that measure asthma exacerbations which use a recall period of "the past month" while the remaining asthma symptoms (coughing a lot, difficulty breathing, or noisy breathing from the chest) and asthma-specific HRQoL items use "the past week".

Children and Adolescents: Percentage (%) change from baseline in Total Nasal Symptoms Score (TNSS) at 4, 16, 52 weeks in participants with concomitant chronic rhinitis

The TNSS is the sum of scores for each of nasal congestion, sneezing, nasal itching, and rhinorrhea using a four-point scale (0-3), where 0 indicates no symptoms, a score of 1 for mild symptoms that are easily tolerated, 2 for awareness of symptoms which are bothersome but tolerable and 3 is reserved for severe symptoms that are hard to tolerate and interfere with daily activity.
TNSS is calculated by adding the score for each of the symptoms to a total out of 12.
