Linhas convergentes de evidências de estudos pós-morte fornecem fortes evidências de que o déficit do receptor muscarínico M1 do cérebro está presente em um subconjunto de pacientes com esquizofrenia (SZ). Os receptores M1 são um alvo importante para déficits cognitivos na SZ. No entanto, até agora, não foi possível examinar a heterogeneidade da SZ em relação à disponibilidade do receptor M1 in vivo. O desenvolvimento de um novo ligante de tomografia por emissão de pósitrons (PET), [11C]EMO, no Yale PET Center oferece uma oportunidade única para, pela primeira vez, examinar in vivo a disponibilidade do receptor muscarínico M1 do cérebro na SZ e, simultaneamente, elucidar a relação dos receptores M1 aos déficits cognitivos na SZ.
Os investigadores irão comparar a disponibilidade do receptor M1 em pacientes com SZ e controles saudáveis pareados por idade e sexo usando [11C]EMO e o High Resolution Research Tomograph (HRRT), um scanner PET com alta sensibilidade e resolução disponível para imagens do cérebro humano. Este estudo explorará a relação entre: ligação hipocampal [11C]EMO (como uma medida da disponibilidade de M1 AChR hipocampal), poder γ relacionado à codificação durante uma tarefa de memória verbal, memória verbal, gênero e nível sérico de acetilcolina. Este estudo exploratório fornecerá os dados piloto necessários para conduzir um estudo maior para investigar completamente a heterogeneidade do SZ em relação à disponibilidade do receptor M1.

Critério de inclusão:
- Homens e mulheres com idade entre 18 e 55 anos que são fisicamente e mentalmente saudáveis, com exceção do DSM-5 esquizofrenia ou diagnóstico de transtorno esquizoafetivo
- Indivíduos sem metal no corpo que possam representar um risco durante a ressonância magnética
- Sem histórico médico significativo, incluindo traumatismo craniano e distúrbios hemorrágicos
Critério de exclusão:
- Homens e mulheres com histórico ou presença de condições médicas clinicamente significativas
- Fumantes que não conseguem se abster do uso do cigarro por 5 dias
- Pessoas que sofrem de claustrofobia, têm implantes incompatíveis com ressonância magnética ou outras contraindicações para exames de ressonância magnética e PET
