Um ensaio clínico multicêntrico de fase 1, aberto, escalonamento e expansão de dose para avaliar a segurança, farmacocinética, farmacodinâmica e eficácia preliminar de ZE50-0134 em leucemia linfocítica crônica recidivante e refratária, linfoma linfocítico pequeno e baixa graduação selecionada Linfomas. A Parte 1 utilizará um design 3+3. Os pacientes elegíveis serão inscritos sequencialmente em cada uma das 5 coortes planejadas de nível de dose. O objetivo do escalonamento da dose da Parte 1 é determinar a dose máxima tolerada de ZE50-0134.

Critérios de inclusão:

  1. Homens e mulheres com idade ≥18 anos.
  2. Doença conforme definida abaixo:

    Parte 1: Pacientes com LLC ou SLL sintomática (definida por iwCLL) sem del(17p)/TP53 devem ter recebido ≥2 terapias anteriores que incluíram BTKi e venetoclax (ou recusaram) ou Pacientes com linfoma progressivo de baixo grau que inclui linfoma de zona marginal, linfoma linfoplasmocitário (incluindo macroglobulinemia de Waldenstrom) que receberam pelo menos 2 terapias, incluindo um anticorpo baseado em BTKi e CD20 terapia.

Lonitoclax is supplied as immediate-release white opaque soft gelatin capsules in 25 mg, 100 mg, and 250 mg strengths. Participants receive a 3-day step-up regimen followed by the assigned oral dose once daily (QD) or twice daily (BID) in a fed state, administered within 1 hour of food. Each cycle is 28 days. The selected regimen may be once daily or twice daily. Approximately 15 participants are planned.
