Flow-medieret dilatation (FMD) - Forsøgspersoner vil ligge i liggende stilling i 20 minutter for at opnå hæmodynamisk steady state. En blodtryksmanchet (Hokanson) vil blive placeret rundt om underarmen (distalt i forhold til Doppler-transduceren) og hurtigt oppustet til 250 mmHg i 5 minutter (cirkulationsstop). Samtidige ultralydsbilleder af karret (B-tilstand) og Doppler-bølgeformer vil blive opsamlet 10 sekunder før og i 2 minutter efter udtømning af manchetten. Alle billeder i B-tilstand vil blive analyseret ved hjælp af automatiseret kantdetektionssoftware (Medical Imaging Applications), mens intensitetsvægtede hastighedsspektresegmenter vil blive gemt på GE Logiq 7-harddisken til off-line blodhastighedsbølgeformsanalyse. P.I. har brugt den traditionelle metode til brachial arterie flow-medieret dilatation (FMD) induceret af reaktiv hyperæmi til at vurdere vaskulær endotelfunktion i populationer lige fra unge raske voksne til ældre voksne med patologiske tilstande.
Spygmocor - Pulse Wave Velocity (PWV) - En Spygmocor®-enhed vil blive brugt ved baseline og efter hver protokol til at vurdere PWV. PWV-analyse giver en ikke-invasiv vurdering af arteriel stivhed. Øget arteriel stivhed er kendt for at være forbundet med hjerte-kar-sygdomme. Deltageren skal ligge i hvilestilling i cirka 30-45 minutter. Forskningsassistenten vil placere EKG-elektrodesensorer på carotis, femoral, radial og distale arterieplaceringer. En meget følsom pen-lignende enhed, kaldet et tonometer, påføres derefter forsigtigt for at registrere hastigheden af blodstrømmen mellem hvert af punkterne.

- Patienter med KOL (GOLD stadier II-IV) og matchede raske kontroller
- Kaukasisk eller afroamerikansk
- Både mænd og kvinder
- Nuværende og tidligere rygere
Ekskluderingskriterier:
- GULD Stage I
- Klinisk diagnose af hjertesygdomme, hypertension eller metabolisk sygdom
- Vasoaktiv medicin (dvs. nitrater, betablokkere, ACE-hæmmere, Viagra osv.)
- Pulmonal hypertension
- Hypothyroidisme
- Hyper-homocysteinæmi
- Interstitiel lungesygdom
- Fenylketonuri
- Graviditet
- Søvnapnø
- Anæmi
- Raynods fænomen
- Koldbrand af cifrene
- Anamnese med lave blodplader eller koagulopatier

EKSPERIMENTEL: KOL-patienter
Patienter med KOL AOC protokol: Brachial arterie flow-medieret dilatation, direkte vurdering af oxidativ stress via EPR spektroskopi (O2-) og biomarkører for oxidativ stress (8-isoprostan, LH, SOD) vil blive vurderet ved baseline og 2 timer efter indtagelse af en enkelt oral oral antioxidantcocktail (1 g vitamin C, 600 IE vitamin E og 600 mg alfa-liponsyre.)
