Explants de foie humain pour HIF-1 Alpha - Analyse/Comparaison

Description détaillée
Dans l'aspect rétrospectif actuellement en cours de l'étude, nous obtiendrons des échantillons d'explants de tumeurs pathologiques de la population d'explants de tumeurs du 1/8/1994 au 31/12/2005. Nous préparerons 5 à 10 lames de chaque bloc d'explants tumoraux et mesurerons HIF-1α par coloration immunohistochimique. Nous effectuerons également un examen rétrospectif des dossiers pour des points de données spécifiques que nous pensons nécessaires pour nous aider dans notre analyse des échantillons histopathologiques.
De plus, nous examinerons les données de la base de données United Network for Organ Sharing (UNOS) : www.unos.org, ce qui nous permettra de développer une analyse plus approfondie et plus robuste de l'expérience du sujet avec le cancer du foie. Il s’agit d’un véhicule de déclaration public et obligatoire auquel le gouvernement oblige tous les centres de transplantation à se rapporter. Même si le sujet a été transplanté à Northwestern et déplacé, l'accès aux données de l'UNOS nous permettra de voir quelle est la morbidité/mortalité de ce patient.
Deuxièmement, nous commencerons à recruter des sujets de manière prospective. Ceux qui ont été traités, ceux qui suivent actuellement un traitement ou ceux qui viennent d’être diagnostiqués. De ces sujets, nous obtiendrons un consentement éclairé pour : a) examiner les dossiers médicaux (données actuelles, futures et rétrospectives), b) collecter des échantillons de sang pour une analyse future et une corrélation avec les données de leurs lames d'explant, c) nous permettre de suivre ces sujets indéfiniment pour obtenir des données sur les résultats en cours, la morbidité et la mortalité. information.

Critère d'intégration:
- Adultes, tout sexe, ≥ à 18 ans
- Diagnostic antérieur ou actuel de carcinome hépatocellulaire (prouvé cliniquement ou par biopsie)
- Résection pour hépatome et/ou greffe
- Patients avec un diagnostic de carcinome hépatocellulaire du 1/8/94 au 31/12/05 (rétrospectif) avec biopsie, explant et/ou transplantation hépatique ici au Northwestern Memorial Hospital (NMH).
- Les patients précédemment diagnostiqués ou récemment diagnostiqués avec un cancer du foie qui ont été traités, sont actuellement traités et subiront potentiellement un traitement pour la maladie.

Une prise de sang en deux temps : une avant le traitement du cancer, la seconde après le traitement du cancer.

Biomarqueur HIF-1alpha

L'objectif principal est de comparer les niveaux d'expression de HIF-1alpha dans les explants de tumeurs CHC qui ont reçu : 1) aucune embolisation pré-explantante ; 2) chimioembolisation artérielle transcathéter pré-explantante (TACE); 3) radio-embolisation Y90 pré-explantante ; 4) ablation par radiofréquence pré-explantante, ou 5) une combinaison de thérapies pré-explantantes.
