Criterio di inclusione:

  • HNSCC incurabile, definito come malattia RM non suscettibile di cura mediante intervento chirurgico e/o radioterapia o paziente con declino HNSCC o non idoneo alla terapia curativa

    • Nella fase I, cavità orale, orofaringe, laringe, ipofaringe, rinofaringe, seno paranasale o ghiandola salivare
    • Nella fase II, cavità orale, orofaringe, laringe o ipofaringe
  • Valutazione della malattia:

    • Nella fase I, malattia valutabile o misurabile.
    • Nella fase II, malattia misurabile secondo RECIST 1.1
  • Trattamento precedente:

    • Per la fase I, qualsiasi numero di linee di terapia precedente per RM-HNSCC.
    • Per la fase II, nessuna precedente terapia sistemica per RM-HNSCC.
  • Almeno 18 anni di età.

Criteri di esclusione:

  • Perforazione o fistola gastrointestinale entro 6 mesi dalla prima dose della terapia del protocollo
  • Storia di problemi gastrointestinali come malattia infiammatoria intestinale, colite ulcerosa o morbo di Crohn.
  • Ipertensione scarsamente controllata (definita come misurazioni della pressione alta [pressioni sistoliche ≥ 160 mmHg o pressioni diastoliche > 100 mmHg] documentate durante l'intervallo di due settimane prima dell'arruolamento). Participants enrolled in Phase I Dose Level 1 received 10 mg/kg ramucirumab administered intravenously (IV) on Day 1. Pembrolizumab was administered as per standard of care (SOC) IV at a dose of 200 mg on Day 1. Each cycle was 21 days.
