Inclusiecriteria:

  • Proefpersonen bij wie onlangs de diagnose symptomatisch multipel myeloom (MM) is gesteld en die:

    • geen eerdere systemische antimyeloomtherapie hebben gekregen EN
    • meetbare ziekte hebben EN
    • komen niet in aanmerking voor een hoge dosis therapie plus stamceltransplantatie (SCT) vanwege leeftijd (≥ 65 jaar) of naast elkaar bestaande aandoeningen. Weigering om een ​​hoge dosis therapie met SCT te ondergaan is NIET voldoende voor deelname aan CA204006 voor een proefpersoon < 65 jaar oud. Er moet een comorbiditeit zijn die SCT verhindert bij een proefpersoon < 65 jaar

Uitsluitingscriteria:

  • Proefpersonen met niet-secretoir of oligo-secretoir myeloom of myeloom met alleen de lichte keten
  • Smeulende MM, gedefinieerd als asymptomatische MM zonder lytische botlaesies

Tijdsspanne: Van randomisatie tot de datum van eerste gedocumenteerde tumorprogressie of overlijden door welke oorzaak dan ook (tot 8 jaar)

PFS wordt gedefinieerd als de tijd vanaf randomisatie tot de datum van de eerste gedocumenteerde tumorprogressie (zoals bepaald door de Independent Review Committee (IRC)) of overlijden door welke oorzaak dan ook.

Het IRC voerde een geblindeerde, onafhankelijke beoordeling uit van de tumorbeoordelingen op basis van de criteria van de European Group for Blood and Bone Marrow Transplant (EBMT).

Toegepaste censureringsregels:

  • Deelnemers die daaropvolgende systemische antimyeloomtherapie kregen voorafgaand aan gedocumenteerde progressie, werden gecensureerd op de datum van de laatste adequate tumorbeoordeling voorafgaand aan nieuwe therapie.
  • Deelnemers met een gebeurtenis (progressie of overlijden)> 10 weken na hun laatste tumorbeoordeling werden gecensureerd bij hun laatste adequate tumorbeoordeling voorafgaand aan het evenement.
  • Deelnemers zonder progressie of overlijden (en die geen daaropvolgende therapie kregen voorafgaand aan progressie) werden gecensureerd bij hun laatste adequate tumorbeoordeling.
  • Deelnemers zonder tumorbeoordelingen na baseline werden gecensureerd op de datum van randomisatie

ORR wordt gedefinieerd als het percentage deelnemers met objectieve respons onder alle gerandomiseerde proefpersonen. Deelnemers met een objectieve respons zijn die deelnemers die een gedeeltelijke respons (PR) of beter ervaren, gebaseerd op de beoordeling van de Independent Review Committee (IRC), volgens de EBMT-criteria.

Overleving wordt gedefinieerd als de tijd vanaf randomisatie tot de datum van overlijden. Van een deelnemer die niet stierf, werd zijn of haar overlevingsduur gecensureerd op de datum van het laatste contact ('laatst bekende datum in leven').

Tijdsspanne: Van baseline tot einde van de behandeling (ongeveer 8 jaar)

Pijnernst (sensorische dimensie) en pijninterferentie (reactieve dimensie, beoordelen van de mate waarin pijn het functioneren verstoort) worden gemeten met behulp van de Brief Pain Inventory-Short Form (BPI-SF).

BPI-SF numerieke beoordelingsschaal gaat van 0 (geen pijn) tot 10 (pijn zo erg als je je kunt voorstellen).
Van baseline tot einde van de behandeling (ongeveer 8 jaar)
Progressievrije overlevingsfrequentie (PFS) op specifieke tijdstippen
Tijdsspanne: Van randomisatie tot het opgegeven tijdstip (tot 5 jaar)
PFS-snelheid wordt gedefinieerd als het percentage deelnemers dat PFS ervaart op de gedefinieerde tijdstippen.
