Dit is een multi-site, gerandomiseerde, gecontroleerde, parallelle voedingsinterventiestudie die is ontworpen om de impact van pulsverrijkte voedingsmiddelen op de cognitieve functie en cardiometabolische gezondheid bij volwassenen met obesitas te onderzoeken. Werving zal bestaan uit ongeveer 160 deelnemers (n= 80 deelnemers per locatie) op 2 locaties in Winnipeg en Zuid-Australië.
Deelnemers worden willekeurig toegewezen aan een van de 2 groepen; een pulsgroep of een controlegroep. De pulsgroep zal met puls verrijkt voedsel consumeren dat ontworpen is om gedurende 12 weken ½ kopje peulvruchten per dag af te geven. De controlegroep consumeert gedurende 12 weken vergelijkende voedingsmiddelen. De puls- en vergelijkende voedingsmiddelen worden aan de deelnemers verstrekt en hen wordt gevraagd deze in hun gebruikelijke dieet op te nemen. Deelnemers wordt verzocht hun eetgewoonten of lichaamsbeweging tijdens de studieperiode niet te veranderen, behalve als dit nodig is om te voldoen aan de studievereisten. Vijfdaagse gewogen voedselrecords zullen worden ingevuld in de week voorafgaand aan de basislijn en tijdens de laatste week van het onderzoek om de inname via de voeding, in het bijzonder de inname van peulvruchten, te controleren. Dit zal bepalen of deelnemers de studievoeding naar behoefte opnemen en zal worden gebruikt om de bijbehorende veranderingen in de inname via de voeding en het lichaamsgewicht te evalueren, indien deze zich voordoen.

Inclusiecriteria:
- Man of vrouw, tussen de 50 en 80 jaar oud
- Body Mass Index (BMI) van >25 kg/m2
- Moet de afgelopen 6 maanden een stabiel regime hebben als u medicijnen of supplementen gebruikt voor verhoogde lipiden, bloeddruk, glucoseregulatie, stemming, pijn of slapeloosheid
- Consumeer 1 of minder porties peulvruchten per week
- Engels kunnen lezen en begrijpen
- Bereid om te voldoen aan de protocolvereisten
- Bereid om geïnformeerde toestemming te geven.
Uitsluitingscriteria:
- Lichaamsgewicht ≥135 kg (aangezien dit groter is dan de mogelijkheid voor Dual Energy X-ray Absorptiometry, d.w.z. DEXA-scanning om de lichaamssamenstelling te beoordelen)
- Gevestigde cardiovasculaire, lever- of nierziekte
- Ongecontroleerde diabetes (hemoglobine HbA1c >8)
- Een score van ≤23 op het Mini Mental State Examination
- Gebruik van eetlustremmers of Orlistat (Xenical)
- Onvermogen om met pulsen verrijkt voedsel te consumeren (d.w.z. bonen, erwten, kikkererwten en linzen) als gevolg van allergieën of ernstige gastro-intestinale reacties
- Zwangerschap
- Roken of pas onlangs gestopt met roken (heeft in de afgelopen 12 maanden sigaretten gerookt)
- Gewichtsverlies van ≥3 kg lichaamsgewicht binnen de 6 maanden voorafgaand aan deelname aan het onderzoek
- Naleving van een dieet voor gewichtsverlies of een programma voor lichaamsbeweging dat is ontworpen om gewichtsverlies te vergemakkelijken
- Acute of terminale ziekte.

Vasculaire functie

|
De vasculaire functie zal niet-invasief worden beoordeeld (bij aanvang, week 6 en week 12) met meting van arteriële compliantie (elasticiteit) door middel van pulsgolfanalyse en pulsgolfsnelheid.
Meting van arteriële vasodilatatie zal niet-invasief worden gedaan om endotheelcellen te evalueren die de bloedvaten bekleden die de bloedstroom reguleren.
Er zullen bloedmonsters worden genomen om markers van vasculaire functie te beoordelen.

Cerebrale bloedstroomsnelheid (alleen Australische site)

De cerebrale bloedstroomsnelheid zal worden beoordeeld (basislijn, week 6, week 12) door middel van transcraniële Doppler-echografie.
Dit zal dienen als een index van de reactiviteit van de arteriële vasodilatator in de cerebrale circulatie.
Dit wordt alleen door de Australische site uitgevoerd.
