1.1 Lactobacillus paracasei subsp. paracasei NTU 101-stam (NTU 101) is een lokale Lactobacillus-stam die werd geïsoleerd en geselecteerd uit de ingewanden van pasgeborenen in Taiwan en die een goed aanpassingsvermogen, maagzuurresistentie en galzoutresistentie vertoont. Deze soort kan lange tijd in de menselijke darm overleven, de groei van schadelijke bacteriën remmen en meerdere rollen spelen. Studies hebben aangetoond dat de NTU 101-stam, of zijn gefermenteerde producten, bijdragen aan verschillende functies, namelijk: (1) het verbeteren van de functie van de darmmicroflora door te voorkomen dat schadelijke bacteriën de darm koloniseren, waardoor ze beschermende effecten uitoefenen; (2) het reguleren van het immuunsysteem door celactivering en proliferatie te stimuleren en de secretie van cytokinen en antilichamen te verhogen om aangeboren en verworven immuunresponsen te versterken; (3) herstel van schade aan het maagslijmvlies door remming van acute maagslijmvliesbeschadiging veroorzaakt door pylorusligatie en aangezuurde alcohol, verlaging van de lipideperoxideconcentraties in het bloed en maagslijmvlies, verhoging van de activiteit van superoxide-dismutase en bevordering van de synthese van prostaglandine E2, die beschermt het slijmvlies; en (4) de verbetering van gastro-intestinale functies die bijdragen aan het verlichten van metabole afwijkingen door de gastro-intestinale functie en het metabolisme te verbeteren. Deze studie is verdeeld in twee secties en richt zich op de effecten van Lactobacillus-suppletie op gezonde mensen die NTU 101 gebruiken. Onderzoekers voerden klinische onderzoeken uit om de effecten te beoordelen van de Vigiis 101-LAB-capsule I op het verbeteren van de gastro-intestinale functie en het reguleren van de darmflora (klinische proef 1), en de Vigiis 101-LAB-capsule II op de verbetering van de peristaltiek, immuniteit en antioxidatieve werking. capaciteit (klinische proef 2).
1.2 Materialen De stam die in de huidige studie werd gebruikt was L. paracasei subsp. paracasei NTU 101 (gevriesdroogd poeder, Vigiis 101-LAB; probiotisch poeder van SunWay Biotech Co., Ltd., Taipei, Taiwan). De Vigiis 101-LAB gemengde lactose, kristallijne cellulose en hulpstof werden tot capsules gemaakt (Vigiis 101-LAB capsule I) met 5 miljard bacteriën per capsule voor de darmflora klinische proef 1. De Vigiis 101-LAB gemengde lactose, kristallijne cellulose en hulpstof werden ook gemengd in capsules (Vigiis 101-LAB capsule II) die 10 miljard bacteriën per capsule bevatten voor klinische proef 2 op peristaltiek, immuniteit en anti-oxidatieve capaciteit. Maltodextrine werd als placebo gebruikt.
1.3 Gerandomiseerde, dubbelblinde klinische criteria van effecten van Vigiis 101-LAB capsule I op de darmflora (klinisch onderzoek 1) Vigiis 101-LAB capsule I werd eenmaal per dag oraal toegediend, telkens één capsule. Het hele onderzoek duurde 4 weken en proefpersonen mochten geen gefermenteerde voedingsproducten eten, zoals miso, kimchi, gefermenteerde zuivelproducten, oligosaccharide-bevattende voedingsmiddelen en melkzuurbacteriën-bevattende producten. De proefpersonen vermeden ook het nuttigen van overmatig gasproducerend voedsel (zoals sojabonen en zoete aardappelen) tijdens hun dagelijkse maaltijden en vermeden voedsel dat een opgezette buik kan veroorzaken of de peristaltiek kan bevorderen (zoals melkzuurdranken en oligosaccharidebevattende dranken). Nadat de proef was begonnen, noteerden de proefpersonen hun dagelijkse aantal stoelgangen en vulden ze relevante vragenlijsten in in week 0, 2 en 4. De proefpersonen bezochten de arts eens in de 2 weken om de gastro-intestinale functie en fysiologische kenmerken te controleren. In week 0, 1, 2, 3 en 4 werden fecale monsters van de proefpersonen verzameld en in flessen met een anaëroob verdunningsmiddel gedaan en geschud om de feces en het verdunningsmiddel gelijkmatig te mengen.
1.4 Gerandomiseerde, dubbelblinde klinische criteria voor het evalueren van de effecten van Vigiis 101-LAB capsule II op peristaltiek, immuniteit en anti-oxidatieve capaciteit (klinisch onderzoek 2) Klinisch onderzoek 2 werd uitgevoerd van maart 2017 tot april 2018 aan de Chung Shan Medical University na het verkrijgen van goedkeuring van de IRB van het Taichung Chung Shan Medical University Hospital (IRB-bewijsdocument CHMUH nr.: CS17018), en alle proefpersonen ondertekenden een formulier voor geïnformeerde toestemming. Vigiis 101-LAB capsule II werd eenmaal per dag oraal toegediend, telkens één capsule. Dagelijks werd het dieet van de proefpersonen vanaf de pre-stabiliteit tot de post-stabiliteit periode tot in detail bijgehouden.
1.5 Uitkomstmetingen Fecaal vochtgehalte en pH-waarde testen. Antropometrische metingen omvatten de lichaamslengte (BH), lichaamsgewicht (BW), bloeddruk (BP), middenarmomtrek (MAC), tricepshuidplooi (TSF), middelomtrek (WC) en rompomtrek (RC) en hartslag ( HR) werden gemeten. De body mass index (= BW/BH2, kg/m2) van de proefpersonen werd berekend na het meten van hun BH en BW. Voor bloeddrukmetingen werden de systolische bloeddruk (SBP) en diastolische bloeddruk (DBP) van de rechterarm gemeten. MAC werd gemeten met hetzelfde paar remklauwen door dezelfde getrainde medewerker. Biochemische tests van de nuchtere bloedmonsters werden verzameld in week 0, 4 en 6. De testitems omvatten bloedbiochemische tests, plasma-antioxidantactiviteit (Trolox-equivalent antioxidantcapaciteit, TEAC), thiobarbituurzuurreactieve stoffen (TBARS), glutathionspiegels (GSH), glutathionperoxidase (GSH Px), glutathionreductase (GSH Rd), volbloed tellen en elektrolyten in het bloed. In het laboratorium werden bloedbiochemische tests uitgevoerd.
1.6. Statistische analyse Gegevens worden uitgedrukt als het gemiddelde ± standaarddeviatie (SD). De statistische significantie van de biochemische analyses werd bepaald door one-way variantieanalyse (ANOVA) met behulp van de algemene lineaire modelprocedure van de software voor statistische product- en serviceoplossingen (SPSS Institute, Inc., Chicago, IL, VS). Dit werd gevolgd door ANOVA met een gepaarde t-test om het verschil voor en na toediening van monster en placebo te evalueren, terwijl de Student t-test werd gebruikt om het verschil tussen test- en placebogroepen te vergelijken (P ≤ 0,05).

Inclusiecriteria:
- Gezonde volwassenen van ≧ 20 jaar en ≦ 65 jaar.
- Proefpersonen met een normaal gewicht (body mass index: 18,5-24).
- Proefpersonen zonder gastro-intestinale aandoeningen of medicijnen.
Uitsluitingscriteria:
- Zwangere of zogende vrouwen.
- Proefpersonen die allergisch zijn voor Lactobacillus.
- Proefpersonen met chronische gastro-intestinale aandoeningen.
- Proefpersonen die eerder een gastrectomie of maagbypass hebben ondergaan.
- Onderwerpen met lever-, nier- of hartaandoeningen, alcoholisme of ongecontroleerde diabetes.
- Proefpersonen die in de afgelopen 1 jaar een beroerte, psychiatrische ziekte of depressie hebben ontwikkeld.
- Onderwerpen die in de afgelopen 2 weken medicijnen hebben gekregen die de gastro-intestinale functie kunnen reguleren, functionele voedingsmiddelen, bacteriostatische medicijnen of supplementen, antibiotica, antioxidanten of andere onbekende medicijnen.
- Patiënten die niet aan de inclusiecriteria voldeden, het testproduct niet volgens de voorschriften toedienden, van wie de werkzaamheid niet kon worden vastgesteld, of die over onvolledige gegevens beschikten die van invloed zijn op de beoordeling van de werkzaamheid of veiligheid.
