Fase I, First-in-Human, Open-Label, Multiple Ascending Dose Study om de veiligheid, verdraagbaarheid, farmacokinetiek, farmacodynamiek en klinische activiteit te onderzoeken van M6223, een remmer van TIGIT, als enkelvoudig middel en in combinatie met Bintrafusp Alfa (anti- PDL1/TGFß Trap) bij deelnemers met gemetastaseerde of lokaal gevorderde vaste inoperabele tumoren
Het hoofddoel van deze studie is het bepalen van de veiligheid, verdraagbaarheid, farmacokinetiek (PK), immunogeniciteit en (indien waargenomen) de maximaal getolereerde dosis (MTD) van M6223 als enkelvoudig middel (Deel 1A) voor zowel de tweewekelijkse (Q2W )-regime en het driewekelijkse (Q3W)-regime en van M6223 gecombineerd met bintrafusp alfa (deel 1B) voor het Q2W-regime bij deelnemers met gemetastaseerde of lokaal gevorderde solide inoperabele tumoren.

Inclusiecriteria:
- Deelnemers hebben histologisch of cytologisch bewezen lokaal gevorderde of gevorderde solide maligniteiten die refractair zijn voor of progressief zijn geworden onder standaardbehandeling en geen andere behandelingsopties hebben waarvan bekend is dat ze klinisch voordeel opleveren
- Deelnemers met Eastern Cooperative Oncology Group Performance Status (ECOG PS) van 0 tot 1 bij screening
- Deelnemer heeft een in formaline gefixeerd, in paraffine ingebed blok met tumorweefsel of minimaal 15 (bij voorkeur 25) ongekleurde tumorglaasjes die geschikt zijn voor op immunohistochemie gebaseerde kleuring van eiwitexpressie
- Deelnemers met een levensverwachting van minimaal 12 weken
- Deelnemers met meetbare ziekte volgens Response Evaluation Criteria in Solid Tumors versie 1.1 (RECIST 1.1)
- Adequate hematologische, lever- en nierfunctie zoals gedefinieerd in het protocol
- Andere in het protocol gedefinieerde opnamecriteria kunnen van toepassing zijn
Uitsluitingscriteria:
- Deelnemers met aanhoudende toxiciteit gerelateerd aan eerdere therapie Graad groter dan (>) 1 National Cancer Institute Common Terminology Criteria for Adverse Events (NCI-CTCAE) versie 5.0, echter alopecia, sensorische neuropathie Graad kleiner dan of gelijk aan (<=) 2, of andere niet-immuungerelateerde Graad <= 2 die geen veiligheidsrisico vormt
- Deelnemers met eerdere orgaantransplantatie inclusief allogene stamceltransplantatie
- Deelnemers met eerdere toxiciteit gerelateerd aan een immuuncontrolepuntremmer Graad groter dan gelijk aan (>=) 3 NCI-CTCAE versie 5.0 tenzij opgelost tot graad <= 1 voorafgaand aan opname in het onderzoek
- Deelnemers met huidige significante cardiale geleidingsafwijkingen, waaronder gecorrigeerd QT-interval (QTcF, gecorrigeerd met Fridericia-formule) verlenging van > 450 milliseconden (ms) op drievoudig 12-afleidingen ECG of verminderde cardiovasculaire functie, ventriculaire tachycardie, hypokaliëmie of een voorgeschiedenis van paroxysmale atriale fibrillatie, ernstige hartritmestoornissen en familiegeschiedenis van plotseling overlijden of lang QT-syndroom
- Een voorgeschiedenis van vasculaire, cardiovasculaire of cerebrovasculaire aandoeningen zoals cerebrovasculair accident/beroerte (minder dan [<] 6 maanden voorafgaand aan inschrijving), myocardinfarct (< 6 maanden voorafgaand aan inschrijving), instabiele angina pectoris, congestief hartfalen (New York Heart Associatieclassificatie Klasse >= II), diepe veneuze trombose (< 3 maanden voorafgaand aan inschrijving) of pulmonaire trombose/embolie (< 3 maanden voorafgaand aan inschrijving)

Participants received an intravenous (IV) infusion of M6223 at escalated doses every 2 weeks (Q2W) or every 3 weeks (Q3W) on Day 1 of each Cycle (Each cycle is of 14 days) according to the recommendation of the SMC(Safety Monitoring Committee) until the maximum tolerated dose(MTD) has been reached or confirmed disease progression.

Na goedkeuring van een nieuw product of een nieuwe indicatie voor een goedgekeurd product in zowel de VS als de Europese Unie, zullen de onderzoekssponsor en/of zijn gelieerde bedrijven onderzoeksprotocollen, geanonimiseerde patiëntgegevens en gegevens op studieniveau en geredigeerde klinische onderzoeksrapporten delen met gekwalificeerde wetenschappelijke en medische onderzoekers, op verzoek, voor zover nodig voor het uitvoeren van legitiem onderzoek.
