DESTINY Borstrespons HER2-laag Europa

Een prospectieve, niet-interventionele studie (NIS) met trastuzumab Deruxtecan voor patiënten met HER2-low-expressie van inoperabele en/of gemetastaseerde borstkanker vergezeld van een ziekteregister van patiënten behandeld met conventionele chemotherapie (DESTINY Breast Respond HER2-low Europe)
Trastuzumab deruxtecan (T-DXd) is als monotherapie geïndiceerd voor de behandeling van volwassen patiënten met inoperabele of gemetastaseerde HER2-laag-borstkanker die eerder chemotherapie hebben gekregen in de gemetastaseerde setting of een recidief hebben ontwikkeld tijdens of binnen 6 maanden na voltooiing van adjuvante chemotherapie.

Gedetailleerde beschrijving
Deze niet-interventionele studie zal de effectiviteit met T-DXd, de demografische en klinische kenmerken van de patiënten, behandelingspatronen, verdraagbaarheid, behandeling van bijwerkingen (ADR's) en patiëntervaring van T-DXd onderzoeken bij patiënten met HER2-laag inoperabel en/of uitgezaaide borstkanker. Patiënten zullen worden behandeld volgens de voorgestelde indicatieverklaring in de samenvatting van de productkenmerken (SPC). Er zal geen geneesmiddel worden toegediend als onderdeel van deze studie.
Gegevens over conventionele chemotherapie (d.w.z. inclusief maar niet beperkt tot capecitabine, eribuline, gemcitabine, paclitaxel en nab-paclitaxel) zullen ook worden verzameld in een ziekteregistergedeelte van het onderzoek.

Deelname Criteria

Studie Bevolking
Volwassen patiënten met inoperabele of gemetastaseerde HER2-low borstkanker die eerder chemotherapie hebben gekregen in de gemetastaseerde setting of een recidief ontwikkelden tijdens of binnen 6 maanden na voltooiing van adjuvante chemotherapie.
Beschrijving
Inclusiecriteria:
- Volwassen patiënt (leeftijd ≥ 18 jaar) met histologisch of cytologisch bevestigde diagnose van inoperabel en/of mBC
- Gedocumenteerde HER2-lage status (IHC1+, IHC2+/ISH-)
- Patiënten die eerder chemotherapie hebben gekregen in de gemetastaseerde setting of patiënten bij wie de ziekte terugkeert tijdens of binnen 6 maanden na voltooiing van adjuvante chemotherapie
- Beslissing om opnieuw een behandeling met T-DXd of conventionele chemotherapie te starten volgens de keuze van de arts per SmPC
- Schriftelijke en ondertekende geïnformeerde toestemming om deel te nemen aan het onderzoek
Uitsluitingscriteria:
- Zwangerschap of borstvoeding
- Patiënten die op het moment van gegevensverzameling voor deze studie deelnemen aan of hebben deelgenomen aan een interventionele studie die geblindeerd blijft.
Er zijn geen specifieke uitsluitingscriteria gedefinieerd, aangezien patiënten zullen worden behandeld volgens de voorgestelde indicatieverklaringen in de SmPC.

Study Group 1
Adult participants with 2L+ HER2-low expressing unresectable and/or metastatic breast cancer who have received prior chemotherapy in the metastatic setting or developed disease recurrence during or within 6 months of completing adjuvant chemotherapy. In addition, patients with HER2-low breast cancer treated with conventional chemotherapy will be enrolled. The participants on conventional chemotherapy will be analyzed as exploratory only. |

Adult participants in the first-line setting with hormone receptor (HR) positive, HER2-low or HER2-(ultra)low breast cancer who have received at least one endocrine therapy in the metastatic setting and who are not considered suitable for endocrine therapy as the next line of treatment and have not received prior chemotherapy for advanced metastatic breast cancer.

The PGI-TT is designed to assess the treatment tolerability of the medicine.
Participants will rate the bother associated with any treatment-related symptoms based on a 5-point scale ranging from 1 (not at all) to 5 (very much).

European Organization for Research and Treatment of Cancer Quality of Life Questionnaire Core 30 (EORTC QLQ-C30) Score in Participants With Unresectable and/or Metastatic Breast Cancer

The EORTC QLQ-C30 questionnaire is designed to measure cancer patients' physical, psychological and social functions.
The EORTC QLQ-C30 scales and single-item scales range in score from 0 to 100.
A higher score for the functioning scales and global health status denotes a better level of functioning (i.e. a better state of the patient), while higher scores on the symptom and single-item scales indicate a higher level of symptoms (i.e. a worse state of the patient).
