Metabolisme av deuterert NNN hos røykfrie tobakksbrukere

Femti sunne røykfrie tobakksbrukere vil bli rekruttert ved å annonsere i Minneapolis-St. Paul-området. Denne forskningen vil bli utført ved Tobacco Research Center ved University of Minnesota. Vanlige røykfrie tobakksbrukere vil bli kontrollert over telefon. Hvis de er kvalifisert etter telefonskjermen, vil deltakerne bli invitert til å delta på et orienteringsbesøk i klinikken hvor samtykke vil bli innhentet og forsøkspersoner vil fylle ut skjemaer om tobakksbruk og medisinsk historie. Forsøkspersoner som er fast bestemt på å være kvalifisert etter orienteringsbesøket, vil returnere til klinikken for et baseline-besøk hvor de vil motta det røykfrie tobakksproduktet og bli instruert om å kun bruke studieproduktet til de kommer tilbake for sin første laboratorieøkt syv dager senere.
Før hver av de tre laboratorieøktene vil forsøkspersonene samle opp all urinen i 24 timer frem til den planlagte timen. På hver laboratorieøkt vil deltakerne bruke studieproduktet og samle spytt- og tobakksjuice med jevne mellomrom. Deltakerne vil også tørke innsiden av kinnet for celler og få blodtappet.
Innsamlede prøver vil bli brukt til å karakterisere metabolsk aktivering og DNA-binding av NNN.

Inklusjonskriterier:
- Vanlig røykfri tobakksbruker
- God fysisk helse uten ustabile medisinske tilstander
- Stabil og god mental helse, dvs. opplever for tiden ikke ustabil eller ubehandlet psykiatrisk diagnose, inkludert rusmisbruk, som bestemt av DSM-IV-kriteriene, i løpet av de siste seks månedene
- Villig til å utføre studieaktiviteter som å få tatt blodprøve, spyttoppsamling, urinsamling, flere klinikkbesøk
- For kvinnelige forsøkspersoner i fertil alder, som ikke er kjent for å være gravide eller ammende, eller planlegger å bli gravide innen de neste 12 månedene
- Gi skriftlig informert samtykke
Ekskluderingskriterier:
- Bruk av andre produkter som inneholder tobakk eller nikotin
- Ustabil medisinsk tilstand
- Tar for tiden noen medisiner som påvirker relevante metabolske enzymer
- Ikke villig til å bruke bytte røykfri tobakksmerke for å bruke tobakk beriket med [pyridin-D4](S)-NNN i løpet av denne studien
