Descrizione
Criterio di inclusione:
- È necessario ottenere un consenso informato scritto prima di eseguire qualsiasi valutazione correlata allo studio.
Uomini e donne tra i 18 e gli 80 anni compresi; le donne in età fertile (definite come quelle che non sono in post-menopausa o sterilizzazione post-chirurgica) devono soddisfare entrambi i seguenti criteri:
- Due test di gravidanza negativi (allo screening e alla randomizzazione, prima della somministrazione)
- Uso di dispositivo intrauterino, da almeno 3 mesi prima dello screening fino ad almeno 4 mesi dopo l'ultima dose di bimagrumab/placebo e.v., e un metodo contraccettivo aggiuntivo (barriera)
- Indice di massa corporea (BMI) ≥ 30 o BMI ≥ 27 con una o più comorbilità associate all'obesità (ad es. ipertensione, insulino-resistenza, apnea notturna o dislipidemia)
- Peso corporeo stabile (± 5 kg) entro 90 giorni dallo screening e peso corporeo
- Avere una storia di almeno uno sforzo comportamentale non riuscito auto-riferito per perdere peso corporeo
- In grado di comunicare bene con lo sperimentatore, rispettare i requisiti dello studio e aderire alla dieta e ai programmi di attività per la durata dello studio
Criteri di esclusione:
- Storia di, o nota ipersensibilità a, farmaci anticorpali monoclonali o una controindicazione a semaglutide (Ozempic® o Wegovy®)
- Uso di altri farmaci sperimentali al momento dell'arruolamento o entro 30 giorni o 5 emivite dall'arruolamento, a seconda di quale sia più lungo o più lungo se richiesto dalle normative locali
- Trattamento con qualsiasi farmaco per l'indicazione dell'obesità negli ultimi 30 giorni prima dello screening
- Diagnosi di diabete (ad esempio, HbA1c ≥ 6,5%) che richiede l'uso corrente di qualsiasi farmaco antidiabetico Nota: la sindrome metabolica non è un'esclusione, anche se gestita con un farmaco antidiabetico come metformina o un inibitore SGLT2. Una diagnosi di prediabete o ridotta tolleranza al glucosio gestita esclusivamente con approcci non farmacologici (ad esempio, dieta ed esercizio fisico) non è un'esclusione.
- Qualsiasi infezione cronica che possa interferire con la condotta o l'interpretazione dello studio come l'epatite B (HBV), l'epatite C (HCV) o il virus dell'immunodeficienza umana (HIV). La storia di epatite A o epatite C trattata con successo non è esclusa. Infezione attiva da COVID-19.
- Donazione o perdita di 400 ml o più di sangue entro 8 settimane prima della somministrazione iniziale, o più a lungo se richiesto dalla normativa locale, o donazione di plasma (> 250 ml) entro 14 giorni prima della prima dose
- Qualsiasi disturbo, riluttanza o incapacità non coperti da nessuno degli altri criteri di esclusione, che a giudizio dello Sperimentatore potrebbero mettere a repentaglio la sicurezza del soggetto o il rispetto del protocollo

The Physical-Functioning domain assesses limitations due to health "now" and consists of 10 items, each rated on a 3-point Likert scale.
Scoring of the domain is norm-based and presented in the form of T-scores, with a mean of 50 and standard deviation of 10; higher scores indicate better levels of function.
