Opis
Kryteria przyjęcia:
- Przed przeprowadzeniem jakichkolwiek ocen związanych z badaniem należy uzyskać pisemną świadomą zgodę.
Mężczyźni i kobiety w wieku od 18 do 80 lat włącznie; kobiety w wieku rozrodczym (określane jako te, które nie są po menopauzie lub sterylizacji pooperacyjnej) muszą spełniać oba poniższe kryteria:
- Dwa negatywne testy ciążowe (podczas badania przesiewowego i podczas randomizacji, przed podaniem dawki)
- Stosowanie wkładki wewnątrzmacicznej od co najmniej 3 miesięcy przed skriningiem do co najmniej 4 miesięcy po ostatniej dawce bimagrumabu/placebo i.v. oraz dodatkowa metoda antykoncepcji (mechaniczna)
- Wskaźnik masy ciała (BMI) ≥ 30 lub BMI ≥ 27 i jedna lub więcej chorób współistniejących związanych z otyłością (np. nadciśnienie, insulinooporność, bezdech senny lub dyslipidemia)
- Stabilna masa ciała (± 5 kg) w ciągu 90 dni od badania przesiewowego oraz masa ciała
- Mieć historię co najmniej jednego nieudanego behawioralnego wysiłku, aby schudnąć
- Potrafi dobrze komunikować się z badaczem, przestrzegać wymagań badania oraz przestrzegać diety i programów aktywności przez cały czas trwania badania
Kryteria wyłączenia:
- Historia lub znana nadwrażliwość na przeciwciała monoklonalne lub przeciwwskazanie do semaglutydu (Ozempic® lub Wegovy®)
- Stosowanie innych leków eksperymentalnych w momencie rejestracji lub w ciągu 30 dni lub 5 okresów półtrwania rejestracji, w zależności od tego, który okres jest dłuższy lub dłuższy, jeśli wymagają tego lokalne przepisy
- Leczenie jakimkolwiek lekiem we wskazaniu otyłości w ciągu ostatnich 30 dni przed badaniem przesiewowym
- Rozpoznanie cukrzycy (np. HbA1c ≥ 6,5%) wymagające bieżącego stosowania jakiegokolwiek leku przeciwcukrzycowego. Uwaga: Zespół metaboliczny nie jest wykluczony, nawet jeśli jest leczony lekiem przeciwcukrzycowym, takim jak metformina lub inhibitor SGLT2. Rozpoznanie stanu przedcukrzycowego lub upośledzonej tolerancji glukozy leczonych wyłącznie metodami niefarmakologicznymi (np. dietą i ćwiczeniami fizycznymi) nie wyklucza.
- Wszelkie przewlekłe infekcje, które mogą zakłócać przebieg badania lub jego interpretację, takie jak wirusowe zapalenie wątroby typu B (HBV), wirusowe zapalenie wątroby typu C (HCV) lub ludzki wirus niedoboru odporności (HIV). Historia skutecznego leczenia wirusowego zapalenia wątroby typu A lub zapalenia wątroby typu C nie jest wykluczona. Aktywna infekcja COVID-19.
- Oddanie lub utrata 400 ml lub więcej krwi w ciągu 8 tygodni przed podaniem pierwszej dawki lub dłużej, jeśli wymagają tego lokalne przepisy, lub oddanie osocza (> 250 ml) w ciągu 14 dni przed podaniem pierwszej dawki
- Wszelkie zaburzenia, niechęć lub niezdolność nieobjęte żadnymi innymi kryteriami wykluczenia, które w opinii badacza mogą zagrozić bezpieczeństwu badanego lub przestrzeganiu protokołu
