Beschreibung
Einschlusskriterien:
- Vor der Durchführung studienbezogener Beurteilungen muss eine schriftliche Einverständniserklärung eingeholt werden.
Männer und Frauen zwischen 18 und 80 Jahren einschließlich; Frauen im gebärfähigen Alter (definiert als diejenigen, die nicht postmenopausal oder postoperativ sterilisiert sind) müssen beide der folgenden Kriterien erfüllen:
- Zwei negative Schwangerschaftstests (beim Screening und bei der Randomisierung, vor der Verabreichung)
- Anwendung eines Intrauterinpessars von mindestens 3 Monaten vor dem Screening bis mindestens 4 Monate nach der letzten Dosis von Bimagrumab/Placebo i.v. und einer zusätzlichen Verhütungsmethode (Barrieremethode).
- Body-Mass-Index (BMI) ≥ 30 oder BMI ≥ 27 mit einer oder mehreren mit Fettleibigkeit assoziierten Komorbiditäten (z. B. Bluthochdruck, Insulinresistenz, Schlafapnoe oder Dyslipidämie)
- Stabiles Körpergewicht (± 5 kg) innerhalb von 90 Tagen nach dem Screening und Körpergewicht
- Haben Sie eine Vorgeschichte von mindestens einer selbstberichteten erfolglosen Verhaltensbemühung, um Körpergewicht zu verlieren
- Kann gut mit dem Ermittler kommunizieren, die Studienanforderungen erfüllen und sich während der Studiendauer an die Diät- und Aktivitätsprogramme halten
Ausschlusskriterien:
- Vorgeschichte oder bekannte Überempfindlichkeit gegen monoklonale Antikörper oder eine Kontraindikation für Semaglutid (Ozempic® oder Wegovy®)
- Verwendung anderer Prüfpräparate zum Zeitpunkt der Registrierung oder innerhalb von 30 Tagen oder 5 Halbwertszeiten nach Registrierung, je nachdem, welcher Zeitraum länger ist, oder länger, wenn dies gemäß den örtlichen Vorschriften erforderlich ist
- Behandlung mit Medikamenten für die Indikation Adipositas innerhalb der letzten 30 Tage vor dem Screening
- Diagnose von Diabetes (z. B. HbA1c ≥ 6,5 %), die die aktuelle Anwendung eines Antidiabetikums erfordert Hinweis: Metabolisches Syndrom ist kein Ausschluss, auch wenn es mit einem Antidiabetikum wie Metformin oder einem SGLT2-Hemmer behandelt wird. Eine Diagnose von Prädiabetes oder eingeschränkter Glukosetoleranz, die ausschließlich mit nicht-pharmakologischen Ansätzen (z. B. Ernährung und Bewegung) behandelt wird, ist kein Ausschluss.
- Alle chronischen Infektionen, die die Studiendurchführung oder -interpretation beeinträchtigen könnten, wie Hepatitis B (HBV), Hepatitis C (HCV) oder Human Immunodeficiency Virus (HIV). Die Vorgeschichte einer erfolgreich behandelten Hepatitis A oder Hepatitis C ist kein Ausschlusskriterium. Aktive COVID-19-Infektion.
- Spende oder Verlust von 400 ml oder mehr Blut innerhalb von 8 Wochen vor der ersten Dosis oder länger, wenn dies durch lokale Vorschriften erforderlich ist, oder Plasmaspende (> 250 ml) innerhalb von 14 Tagen vor der ersten Dosis
- Jede Störung, Unwilligkeit oder Unfähigkeit, die nicht durch eines der anderen Ausschlusskriterien abgedeckt ist und die nach Ansicht des Ermittlers die Sicherheit des Probanden oder die Einhaltung des Protokolls gefährden könnten

The SF-36v2 acute form assesses HRQoL on 8 domains: Physical Functioning, Role-Physical, Bodily Pain, General Health, Vitality, Social Functioning, Role-Emotional, and Mental Health.
The Physical-Functioning domain assesses limitations due to health "now" and consists of 10 items, each rated on a 3-point Likert scale.
Scoring of the domain is norm-based and presented in the form of T-scores, with a mean of 50 and standard deviation of 10; higher scores indicate better levels of function.
