Onderzoek ter evaluatie van het effect van Roflumilast 500 μg op het aantal exacerbaties bij proefpersonen met chronische obstructieve longziekte (COPD) die werden behandeld met een vaste-dosiscombinatie van langwerkende bèta-agonist en inhalatiecorticosteroïde (LABA/ICS)
Om het extra voordeel van roflumilast aan te tonen wanneer het wordt toegevoegd aan vaste-dosiscombinatie (FDC) LABA/ICS bij het verminderen van exacerbaties bij personen met ernstige tot zeer ernstige COPD.

Deelname Criteria
Onderzoekers zoeken naar mensen die aan een bepaalde beschrijving voldoen, de zogenaamde geschiktheidscriteria. Enkele voorbeelden van deze criteria zijn iemands algemene gezondheidstoestand of eerdere behandelingen.

Beschrijving
Inclusiecriteria:
- 1. Mannelijke of vrouwelijke patiënten van minstens 40 jaar oud
- 2. Geschiedenis van COPD (volgens Global Initiative for Chronic Obstructive Lung Disease [GOLD] 2010) gedurende ten minste 12 maanden voorafgaand aan de screening (bezoek 1) geassocieerd met chronische productieve hoest gedurende 3 maanden in elk van 2 opeenvolgende jaren (met andere oorzaken van productieve hoest uitgesloten). Alleen patiënten met chronische bronchitis worden opgenomen (gelijktijdig emfyseem is toegestaan)
- 3. Verhouding geforceerd expiratoir volume na 1 seconde (FEV1)/geforceerde vitale capaciteit (FVC) (postbronchodilatator) < 70% bij screening (bezoek 1)
- 4. FEV1 (postbronchodilatator) ≤ 50% van voorspeld bij screening (bezoek 1)
- 5. Ten minste twee gedocumenteerde matige of ernstige COPD-exacerbaties binnen 12 maanden voorafgaand aan de screening (bezoek 1)
- 6. Patiënten moeten ≥ 3 maanden voorafgaand aan de screening een FDC LABA/ICS-behandeling ondergaan (bezoek 1)
- 7. Voormalige rokers (gedefinieerd als stoppen met roken ten minste 1 jaar geleden) of huidige rokers (inclusief patiënten die in het afgelopen jaar zijn gestopt met roken), beide met een rookgeschiedenis van ten minste 20 pakjaren
Uitsluitingscriteria:
- 1. Matige of ernstige COPD-exacerbaties en/of COPD-exacerbaties behandeld met antibiotica of systemische glucocorticosteroïden binnen 4 weken na screening (bezoek 1) (dwz patiënten moeten klinisch stabiel zijn)
- 2. Bekende alfa-1-antitrypsinedeficiëntie
- 3. Huidige diagnose van astma (gecontroleerd of ongecontroleerd) (Opmerking: de geschiedenis van astma bij kinderen sluit niet uit.)
- 4. Lichaamsmassa-index (BMI) ≥ 45 kg/m2
- 5. Patiënten met een voorgeschiedenis (binnen 5 jaar) of huidige diagnose van andere kanker dan basale of plaveiselcelkanker
