Een single-arm, open-label, multicenter klinisch onderzoek met nivolumab (BMS-936558) voor proefpersonen met histologisch bevestigd stadium III (niet-resectabel) of stadium IV melanoom voortschrijdend na eerdere behandeling met een anti-CTLA4 monoklonaal antilichaam
Het doel van deze studie is het bepalen van het aantal en de frequentie van hooggradige (CTCAE v4.0 graad 3 of hoger), behandelingsgerelateerde, geselecteerde bijwerkingen bij personen met histologisch bevestigd stadium III (inoperabel) of stadium IV melanoom en progressie na eerdere behandeling met een anti-cytotoxisch T-lymfocytenantigeen (CTLA-4) monoklonaal antilichaam, behandeld met nivolumab (BMS-936558) in een dosis van 3 mg/kg om de twee weken.

Inclusiecriteria:
- Proefpersonen met histologisch bevestigd kwaadaardig melanoom
Eastern Cooperative Oncology Group (ECOG) Prestatiestatus (PS):
- PS 0 tegen 1
- PS2
- Eerder behandeld inoperabel stadium III- of stadium IV-melanoom volgens de richtlijnen van de American Joint Committee on Cancer 2010, ongeacht de BRAF-mutatiestatus
- Proefpersonen moeten evalueerbare Response Evaluation Criteria In Solid Tumors (RECIST 1.1)-gedefinieerde ziekteprogressie hebben ervaren
- Voorafgaande behandeling met chemotherapie, interferon (adjuvante setting), interleukine (IL-2), BRAF/MEK-remmers voor proefpersonen met bekende BRAF-mutaties, door mitogeen geactiveerde of extracellulaire signaalgereguleerde proteïnekinase (MEK)-remmers voor neuroblastoom Ras Viral (v- ras) oncogene homoloog (NRAS) mutaties, en cKIT-remmers met bekende cKIT-mutaties zijn toegestaan
Patiënten met CZS-metastasen komen in aanmerking:
- als CZS-metastasen worden behandeld, zijn patiënten asymptomatisch of keren neurologisch terug naar de uitgangswaarde
- als ze eerder onbehandelde CZS-metastasen hebben en asymptomatisch zijn
- als ze leptomeningeale metastasen hebben, worden behandeld en asymptomatisch of neurologisch terugkeren naar de uitgangswaarde met een levensverwachting > 3 maanden
- Patiënten met een bekende voorgeschiedenis van graad 3-4 immuungerelateerde bijwerkingen tijdens/na anti-CTLA-4-therapie als alle toxiciteiten ten minste tot graad 1 zijn verdwenen
Uitsluitingscriteria:
- Proefpersonen met onbehandelde, actieve metastasen van het centrale zenuwstelsel (CZS) zijn uitgesloten
