Beschrijving

Inclusiecriteria:

  • Proefpersonen met histologisch bevestigd stadium IV of recidiverende NSCLC plaveiselcel of niet-plaveiselcel histologie, zonder eerdere systemische antikankertherapie
  • Proefpersonen moeten geprogrammeerde death-ligand 1 (PD -L1) immunohistochemische (IHC) testen ondergaan, met resultaten, uitgevoerd door het centrale laboratorium tijdens de screeningperiode
  • Eastern Cooperative Oncology Group (ECOG) Prestatiestatus van ≤ 1
  • Meetbare ziekte door middel van CT of MRI per respons evaluatiecriteria bij solide tumoren versie 1.1 (RECIST 1.1) criteria

Uitsluitingscriteria:

  • Proefpersonen met onbehandelde metastasen van het centrale zenuwstelsel (CZS) zijn uitgesloten
  • Proefpersonen met een actieve, bekende of vermoede auto-immuunziekte zijn uitgesloten
  • Elke positieve test voor hepatitis B-virus of hepatitis C-virus of humaan immunodeficiëntievirus (HIV) wijst op een acute of chronische infectie

Progressievrije overleving (PFS) wordt gedefinieerd als de tijd tussen de datum van randomisatie en de datum van eerste gedocumenteerde ziekteprogressie, gebaseerd op BICR-beoordelingen (volgens RECIST v1.1), of overlijden door welke oorzaak dan ook, afhankelijk van wat zich het eerst voordoet op basis van Kaplan-Meier-schattingen. Progressieve ziekte (PD): Ten minste een 20% toename in de som van diameters van doel laesies, waarbij de kleinste som tijdens de studie als referentie wordt genomen (dit omvat de baseline som als die de kleinste is in de studie). Naast de relatieve toename van 20% moet de som ook een absolute toename van ten minste 5 mm aantonen.

Objectieve responsratio (ORR) wordt gedefinieerd als het aantal deelnemers dat een beste algemene respons (BOR) bereikt van bevestigde volledige respons (CR) of bevestigde gedeeltelijke respons (PR), gebaseerd op geblindeerde onafhankelijke centrale beoordelingen (BICR) met behulp van Response Evaluation Criteria in Solid Tumors (RECIST 1.1), gedeeld door het aantal alle gerandomiseerde deelnemers.

Volledige respons (CR): Verdwijning van alle doellaesies. Elke pathologische lymfeklier (of deze nu doelwit of niet-doelwit is) moet een reductie in de korte as hebben tot < 10 mm.

Gedeeltelijke respons (PR): Ten minste een 30% afname in de som van diameters van doellaesies, waarbij wordt verwezen naar de baseline somdiameters.

De Longkanker Symptoom Schaal (LCSS) is een ziektegerelateerde maatstaf voor kwaliteit van leven die zes belangrijke longkankersymptomen en hun effect op algemene distress en symptoomernst, impact op dagelijkse activiteiten, en algemene kwaliteit van leven evalueert. Deze door patiënt gerapporteerde uitkomst (PRO) vragenlijst beoordeelt de volgende 6 symptoomitems (eetlustverlies, vermoeidheid, hoesten, dyspneu, hemoptoë, pijn) en 3 samenvattende globale items (symptoomdistress, activiteitenniveau, algemene kwaliteit van leven) voor patiënten met behulp van visueel analoge schalen (VAS) (100 mm horizontale lijn) variërend van 0 (beste beoordeling) tot 100 (slechtste beoordeling). De LCSS gemiddelde totalscore is de som van items 1 tot 9 gedeeld door het totale aantal items ((som van items 1 tot 9)/9) variërend van 0 tot 100 waarbij een hoge score de slechtste uitkomst vertegenwoordigt. Ziektegerelateerde symptoomverslechtering tegen week 12 gedefinieerd als een toename van 10 punten of meer ten opzichte van de baseline in de LCSS gemiddelde score op enig moment (tijdens of na de behandeling) tot 95 dagen na de randomisatiedatum.
