Een fase 3, multicenter, gerandomiseerde, dubbelblinde, placebogecontroleerde studie waarin de werkzaamheid en veiligheid van chemotherapie met Golcadomide Plus R-CHOP wordt vergeleken met chemotherapie met Placebo Plus R-CHOP bij deelnemers met eerder onbehandeld hoogrisico groot B-cellymfoom (GOLSEEK) -1)

Het doel van deze studie is om de werkzaamheid en veiligheid van golcadomide in combinatie met rituximab, cyclofosfamide, doxorubicine, vincristine en prednison (R-CHOP) chemotherapie te vergelijken met placebo in combinatie met R-CHOP chemotherapie bij deelnemers met eerder onbehandelde hoogrisicotherapie. groot B-cellymfoom (LBCL).

Inclusiecriteria:

- Histologisch bevestigde (volgens lokale evaluatie) diagnose van de novo, eerder onbehandeld grootcellig B-cellymfoom (LBCL) volgens de classificatie van de Wereldgezondheidsorganisatie (WHO) uit 2022, waaronder:

i) Diffuus grootcellig B-cellymfoom (DLBCL), niet anders gespecificeerd [inclusief typen kiemcentrum B-cellen (GCB) en geactiveerde B-cellen (ABC)]

ii) Hooggradig B-cellymfoom, met MYC- en BCL2-herschikkingen

iii) Hooggradig B-cellymfoom, niet anders gespecificeerd

iv) T-cel/histiocyt/rijk grootcellig B-cellymfoom (THRLBCL)

v) Epstein-Barr-virus + DLBCL

  • International Prognostic Index (IPI) score 1 of 2 met lactaatdehydrogenase (LDH) ≥ 1,3 x bovengrens van normaal (ULN) en/of omvangrijke ziekte gedefinieerd als enkele laesie van ≥ 7 cm OF IPI ≥ 3.
  • Meetbare ziekte gedefinieerd door ten minste 1 fluorodeoxyglucose (FDG)-avid laesie voor FDG-avid subtype en 1 bidimensionaal meetbare (> 1,5 cm in langste diameter) ziekte door computertomografie (CT) of magnetische resonantie beeldvorming (MRI), zoals gedefinieerd volgens de Lugano-classificatie.
  • Moet de ziekte van Ann Arbor stadium II-IV hebben.

Uitsluitingscriteria:

  • Elke significante medische aandoening, actieve infectie, laboratoriumafwijking of psychiatrische aandoening waardoor de deelnemer niet aan het onderzoek zou kunnen deelnemen.
  • Elk ander subtype lymfoom. Gevallen van primair mediastinaal (thymus) grootcellig B-cellymfoom (PMBCL), primair cutaan DLBCL-beentype, graad 3b FL, FL getransformeerd naar a-BCL, anaplastisch lymfoomkinase (ALK) positief grootcellig B-cellymfoom, primair effusielymfoom en Burkitt-lymfoom.
  • Gedocumenteerde of vermoedelijke betrokkenheid van het centrale zenuwstelsel (CZS) door lymfoom.
  • Andere in het protocol gedefinieerde opname-/uitsluitingscriteria zijn van toepassing.
