Deze pilot, gerandomiseerde interventiestudie heeft tot doel het effect te beoordelen van een voedingssupplement dat epigallocatechinegallaat (EGCG), vitamine B12, hyaluronzuur en foliumzuur bevat op het behoud van de fysiologische balans en de natuurlijke afweer van het mannelijke genitale kanaal bij mannen met een HPV-infectie die centra voor natuurlijke voortplanting bezoeken. De studie probeert ook te evalueren of suppletie de spermakwaliteit kan ondersteunen, het reproductieve welzijn van deelnemende paren kan verbeteren en risicofactoren die verband houden met HPV-overdracht naar de vrouwelijke partner kan verminderen.
In totaal zullen 48 paren (96 deelnemers) worden ingeschreven bij het Istituto Scientifico Internazionale "Paolo VI" van het Policlinico A. Gemelli. In aanmerking komende paren, gekenmerkt door een HPV-positieve mannelijke partner en een HPV-negatieve vrouwelijke partner, worden gerandomiseerd in twee groepen: een supplementgroep, waarin beide partners gedurende 6 maanden eenmaal daags orale suppletie krijgen met EGCG (200 mg), vitamine B12 (1 mg), hyaluronzuur (50 mg) en foliumzuur (400 μg), en een controlegroep die geen suppletie krijgt. Randomisatie wordt uitgevoerd met behulp van een computergegenereerde lijst.
Het primaire eindpunt is het aandeel mannelijke deelnemers dat aan het einde van de interventieperiode van 6 maanden een negatieve HPV-DNA-test behaalt. Secundaire eindpunten zijn veranderingen in spermakwaliteitsparameters beoordeeld door sperma-analyse volgens de WHO-richtlijnen van 2021, maatregelen van reproductieve uitkomsten zoals vruchtbaarheidspotentieel en het bereiken van zwangerschap, en het behoud van de gezondheid van het genitale slijmvlies, waaronder afwezigheid van infecties of recidieven en verminderd risico op HPV-overdracht. HPV-genotypering wordt beoordeeld door PCR op urethrale swabmonsters. De totale studieduur wordt verwacht 18 maanden te zijn na goedkeuring door de ethische commissie.
