Descripción detallada

Este es un estudio de fase IIIb, multicéntrico, abierto, aleatorizado de asciminib oral 80 mg una vez al día (QD) versus nilotinib 300 mg dos veces al día (BID) en pacientes adultos con LMC Ph+ en fase crónica recién diagnosticada.

Los participantes serán aleatorizados en el estudio en una proporción de 1:1 a asciminib o nilotinib. No se permitirá el cruce del tratamiento del estudio entre los brazos.

Los participantes serán tratados hasta toxicidad inaceptable, progresión de la enfermedad y/o según el criterio del investigador o de los participantes. Se realizará una visita/llamada de seguimiento de seguridad aproximadamente 30 días después de la visita de finalización del tratamiento. A los participantes que interrumpan el tratamiento del estudio de forma prematura por cualquier motivo, se les hará un seguimiento de supervivencia y progresión (a fase acelerada (AP)/crisis blástica (BC)) hasta el final del estudio.

Criterios de inclusión:

  1. Pacientes con CML-CP dentro de los 3 meses posteriores al diagnóstico.
  2. Diagnóstico de LMC-PC (criterios ELN 2020) con confirmación citogenética del cromosoma Filadelfia

    La LMC en fase crónica documentada cumplirá todos los criterios siguientes Baccarani et al 2013:

    • < 15% de blastos en sangre periférica y médula ósea,
    • < 30% de blastos más promielocitos en sangre periférica y médula ósea,
    • < 20% de basófilos en sangre periférica,

Change from baseline in Overall Scores and individual domains of the EORTC QLQ-C30. The EORTC QLQ-C30 contains 30 items and is composed of both multi-item scales and single-item measures based on the participant's experience over the past week. These include five functional scales (physical, role, emotional, cognitive and social functioning), three symptom scales (fatigue, nausea/vomiting, and pain), six single items (dyspnea, insomnia, appetite loss, constipation, diarrhea and financial impact) and a global health status/QoL scale.

Change from baseline in Overall Scores and individual domains of the EORTC QLQ-CML24. The EORTC QLQ-CML24 assesses specific concepts relevant to the experience of patients with CML. The QLQ-CML24 has 24 items which assess symptom burden, impact on daily life and on worry/mood, body image problems, and satisfaction with care and with social life based on the participant's experience over the past week.
