상세 설명
이 프로젝트는 American Heart Association Go Red for Women Strategically Focused Research Network(SFRN)에서 자금을 지원합니다. 이 SFRN에는 임상 과학 프로젝트(PI, St-Onge)와 기초 과학 프로젝트(PI, Jelic)의 두 가지 개별 프로젝트가 포함됩니다. 여성은 임상 과학 프로젝트에 등록되며 기초 과학 프로젝트에 참여할 수 있는 옵션이 제공됩니다. 각 프로젝트(임상 및 기본)에는 별도의 기본 및 보조 결과가 있습니다. 임상 과학 프로젝트의 경우 주요 결과는 포도당 내성과 보행 및 진료실 혈압입니다. 2차 결과는 체성분 및 순환 염증 표지자입니다. 기초 과학 프로젝트의 경우 일차 결과는 상완 동맥(FMD)의 유동 매개 확장이고 이차 결과는 내피 세포 산화 스트레스 및 염증입니다. 이 두 프로젝트의 결과는 별도의 출판물과 결합된 출판물로 보고될 것입니다.
이 연구는 무작위, 교차, 외래 환자 수면 제한(SR) 연구로 각각 6주씩 2단계로 이루어지며 각 단계 사이에 6주의 휴약 기간이 있습니다. 각 단계의 수면 시간은 습관적 수면(HS) 단계 및 SR 단계에서 HS에서 1.5시간을 뺀 동안 참가자의 정규 취침 및 기상 시간입니다. HS 단계에서 참가자는 스크리닝 수면 일정에 따라 정해진 취침 시간을 따르도록 요청받습니다. SR 단계에서 참가자는 습관적인 기상 시간을 일정하게 유지하되 총 수면 시간을 1.5시간 단축하기 위해 취침 시간을 늦춰야 합니다.
각 연구 단계(기준선)의 첫날에 참가자는 밤새 12시간 금식한 후 아침에 연구 센터에 올 것입니다. 참가자는 인체 측정을 한 다음 방사선과로 이동하여 신체 구성을 평가하기 위해 자기 공명 영상(MRI) 스캔을 받게 됩니다. 유동 매개 확장은 심장혈관 초음파 실험실에서 수행되고 내피 세포 수확(Basic Science 프로젝트)이 뒤따를 것입니다. 내피 기능을 평가하기 위해 혈관(팔뚝 정맥)을 감싸고 있는 세포 샘플을 채취합니다. 여성은 경구 포도당 내성 검사(임상 과학 프로젝트)를 받게 됩니다. 호르몬 측정을 위해 혈액 샘플을 채취합니다(인슐린, 코르티솔). 참가자는 그날 밤 정해진 취침 시간 루틴을 시작합니다. 이러한 기준선 측정은 6주 후 종점에서 반복됩니다. 매주 체중과 허리둘레를 측정하고 순응도 확인 방문 중에 3주와 4주에 공복 혈액 샘플을 채취합니다.
참가자가 수면 프로토콜을 준수하는지 확인하기 위해 수면 일기를 작성하고 하루 24시간 활동 모니터를 착용해야 합니다. 참가자는 손목에 액티그래프 GT3X+를 착용하여 수면 시간, 수면 시간 및 신체 활동을 추적합니다. 무작위 배정 2주 전과 세척 기간 동안 참가자는 또한 액티그래프 시계를 착용하고 수면 일기를 작성하여 수면 시간과 수면 깨우기 일정을 확인합니다. 7-9시간/밤의 평균 수면을 달성하는 것 외에도 손목 액티그래피에 의해 참가자는 14일 중 최소 10일 동안 7시간의 수면을 달성하고 <6시간으로 <4일 밤을 보낸 경우에만 등록됩니다. 수면. 무작위 배정 시 소변 임신 검사와 약물 선별 검사를 실시합니다. 참가자는 시작 전 24시간 동안 카페인과 알코올 섭취를 삼가해야 합니다. 연구 단계 사이에 참가자가 6주의 세척 기간 내에 기본 수면 패턴으로 돌아오지 않으면 추가 2-4주의 세척 기간이 제공됩니다. 이 세척 길이는 여성이 각 실험 단계가 시작될 때 월경 주기의 동일한 단계에 있도록 합니다.
