- 成人(18-75 岁)患有特发性或遗传性 PAH,与结缔组织疾病或使用厌食剂有关。
肺动脉平均压≥25mmHg,肺动脉楔压≤15mmHg。 此外,受试者将需要满足以下血液动力学标准:
- PVR >480 达因.s.cm-5
- 在特发性、遗传性和药物/毒素诱导的 PAH 中(在基线或之前的 RHC 期间)必须进行阴性血管反应性测试。
- 世界卫生组织功能等级 (WHO FC) II 或 III。
- 筛选前 PAH 的适当稳定治疗≥4 个月,包括波生坦和/或 5 型磷酸二酯酶 (PDE-5) 抑制剂和/或前列腺素类以外的内皮素受体拮抗剂 (ERA)。
- 两次 150-550m 的 6 分钟步行测试,彼此之间的误差在 ±15% 以内(后者用作基线值)。

排除标准:
- PAH与HIV感染、门静脉高压症或先天性心脏病有关。
- 由左心疾病(WHO PH 组 2)、肺部疾病和/或缺氧(WHO PH 组 3)、慢性血栓栓塞性肺动脉高压(WHO PH 组 4)或不明的多因素机制(WHO PH 组 5)引起的肺动脉高压。
- 已知或疑似肺静脉闭塞性疾病 (PVOD)。
- 严重限制性肺病(总肺活量 < 60% 预测值)
- 严重阻塞性肺病(支气管扩张剂后 FEV1/FVC < 60%)
- DLCO <40%
- 收缩压 <90 mmHg
- 清醒患者的静息心率 <50 BPM 或 >110 BPM
- 最近 3 个月内临床不稳定的右心衰竭或属于 WHO FC IV。
- 在筛选后 30 天内收到任何研究药物。
- 筛选时体重指数 (BMI) <18 或 >40 kg/m2。
- 患者在接受研究治疗期间以及最后一剂研究治疗后 3 个月内不得怀孕、哺乳或计划怀孕。

筛选时存在 ≥ 3 个射血分数保留的心力衰竭的以下危险因素:

右心衰竭以外的器官功能障碍的证据,包括:
- 肌酐清除率 <45 毫升/分钟(使用 Cockroft-Gault 公式)。
- 血清 AST 或 ALT >3 x ULN。
- 总胆红素 > 1.5 x ULN。
- Childs-Pugh B-C 级肝硬化。
- 血红蛋白 <100 克/升。
- 中性粒细胞绝对计数 < 1,500/μL。
- 血小板 < 150,000/μL。
