Les participants doivent répondre à tous les critères suivants pour être inclus dans l'étude :
Démographie
- Participants adultes masculins ou féminins en bonne santé âgés de 18 à 50 ans (inclus).
Poids corporel entre 50 et 100 kg (inclus) et indice de masse corporelle (IMC) entre 18 et 32 kg/m2 (inclus).
État de santé
En bonne santé tel que déterminé par des tests de dépistage. Une bonne santé est définie comme n'ayant aucune anomalie cliniquement pertinente identifiée par des antécédents médicaux détaillés, un examen physique complet (y compris la mesure de la pression artérielle et du pouls), un ECG à 12 dérivations et des tests de laboratoire clinique.
Signes vitaux (mesurés après 5 minutes de repos en position assise) dans la plage normale, ou en dehors de la plage normale et non considérés comme cliniquement significatifs par l'investigateur.
Paramètres ECG standard à 12 dérivations (enregistrés après un repos de 5 minutes en décubitus dorsal) dans les plages suivantes ; Intervalle QT corrigé (QTc) (algorithme de Fridericia recommandé) ≤ 450 ms pour les hommes et 470 ms pour les femmes, et tracé ECG normal ou tracé ECG anormal non considéré comme cliniquement pertinent par l'investigateur.
Paramètres de laboratoire ne démontrant aucune anomalie cliniquement significative, tels que déterminés par l'investigateur. Une bilirubine totale en dehors de la plage normale peut être acceptable si la bilirubine totale ne dépasse pas 1,5 × la limite supérieure de la bilirubine conjuguée normale (LSN) (à l'exception d'un participant atteint d'un syndrome de Gilbert documenté).
- Un résultat négatif sur le dépistage de drogue dans l'urine et un résultat négatif répété le jour -1 (amphétamines/méthamphétamines, barbituriques, benzodiazépines, cannabinoïdes, cocaïne, opiacés).
Les participantes ne doivent pas être enceintes ni allaiter et doivent utiliser une méthode de contraception efficace (telle que décrite à la section 4.5.4), à l'exception des participantes qui ont subi une stérilisation au cours des 3 mois précédents ou qui sont ménopausées.
Une femme en âge de procréer (WOCBP) doit subir un test de grossesse avant la première dose du médicament expérimental (IMP). La participante doit être exclue de l'étude si le test de grossesse sérique est positif.
Un état postménopausique est défini comme 12 mois d'aménorrhée sans autre cause médicale. En l'absence de 12 mois d'aménorrhée, la ménopause peut être confirmée par la mesure de l'hormone folliculo-stimulante (FSH) (> 40 UI/L ou unité milli-internationale (mUI)/mL). Les femmes sous traitement hormonal substitutif (THS), où la ménopause statut est indéterminé, devra utiliser une méthode contraceptive hormonale sans œstrogène si les participants souhaitent poursuivre leur THS pendant l'étude. Les participantes doivent sinon interrompre le THS pour permettre la confirmation du statut postménopausique avant l'inscription à l'étude.
Régulation
- Fournir un consentement éclairé écrit avant d'entreprendre toute procédure liée à l'étude.
- Ne doit être sous aucune tutelle administrative ou judiciaire ou institutionnalisée en vertu d'un ordre réglementaire ou judiciaire.
Critère d'exclusion:
Les participants qui répondent à l'un des critères suivants seront exclus de l'étude :
Antécédents médicaux et état clinique
- Antécédents ou présence de maladies cardiovasculaires, pulmonaires, gastro-intestinales, hépatiques, rénales, métaboliques, hématologiques, neurologiques, musculo-squelettiques, rhumatologiques, psychiatriques, systémiques, oculaires ou infectieuses cliniquement pertinentes, ou signes de maladie aiguë.
- Maux de tête et/ou migraines sévères fréquents, nausées et/ou vomissements récurrents (définis comme des vomissements plus de deux fois par mois).
- A fait un don de sang de n'importe quel volume dans les 2 mois précédant la première dose.
- Hypotension posturale symptomatique, indépendamment de la diminution réelle de la pression artérielle, ou hypotension posturale asymptomatique avec une diminution de la pression artérielle systolique ≥ 30 mmHg dans les 3 minutes suivant le passage de la position couchée à la position debout.
- Présence ou antécédents d'hypersensibilité médicamenteuse ou de réaction anaphylactique, diagnostiquée et traitée par un médecin.
- Hypersensibilité connue à l'un des composants de la formulation IMP.
- Antécédents ou présence d'abus de drogues ou d'alcool (définis comme la consommation d'alcool de plus de 2 unités par jour sur une base régulière).
- Tabagisme régulier (défini comme plus de 5 cigarettes ou équivalent par semaine), ou incapacité à arrêter de fumer pendant l'étude. Les fumeurs occasionnels peuvent être inscrits.
Consommation excessive de boissons contenant des bases xanthiques (définies comme plus de 4 verres par jour).
Substances interférentes
- Tout médicament, y compris le millepertuis, dans les 14 jours précédant l'administration de la première dose ou dans les 5 fois la demi-vie d'élimination ou la demi-vie pharmacodynamique du médicament, à l'exception de la contraception hormonale, de l'hormonothérapie substitutive de la ménopause ou des paracétamol à des doses allant jusqu'à 2 g/jour.
- Toute consommation de pamplemousse ou de produits contenant du pamplemousse dans les 5 jours précédant l'administration de la première dose.
Toute vaccination dans les 28 jours précédant l'administration de la première dose.
Conditions générales
- Tout participant qui, de l'avis de l'investigateur, est susceptible d'être non conforme pendant l'étude, ou d'être incapable de coopérer en raison de problèmes de langage ou d'un développement mental médiocre.
- Tout participant qui s'est inscrit ou a participé à toute autre étude clinique impliquant un médicament expérimental, ou à tout autre type de recherche médicale dans le mois ou dans les 5 fois la demi-vie d'élimination avant l'administration de la première dose.
- Tout participant non joignable en cas d'urgence.
Tout participant qui est l'investigateur ou tout sous-investigateur, assistant de recherche, pharmacien, coordinateur de l'étude ou autre membre de son personnel directement impliqué dans la conduite de l'étude ou toute personne dépendante (employés ou membres de la famille immédiate) du site de l'étude, de l'investigateur ou du Parrainer.
Statut biologique
- Résultat positif à l'un des tests suivants : antigène de surface de l'hépatite B (HBsAg), anticorps anti-hépatite B (HBcAb), anticorps anti-virus de l'hépatite C (anti-VHC), anticorps anti-virus de l'immunodéficience humaine 1 et 2 (anti-VIH1) et Ac anti-VIH2).
- Test d'alcoolémie positif.
- Tout participant chez qui le prélèvement de sang veineux est difficile.
