Badanie fazy II, z podwójnie ślepą próbą, kontrolowane placebo, randomizowane, z różnymi dawkami, w równoległych grupach, oceniające bezpieczeństwo i skuteczność PHA-022121 podawanego doustnie w profilaktyce przeciw napadom obrzęku naczynioruchowego u pacjentów z dziedzicznym obrzękiem naczynioruchowym spowodowanym niedoborem inhibitora C1 ( Typ I lub Typ II)
Niniejsze badanie ocenia bezpieczeństwo i skuteczność PHA-022121 podawanego doustnie w profilaktyce napadów obrzęku naczynioruchowego u pacjentów z dziedzicznym obrzękiem naczynioruchowym (HAE).
Badanie składa się z 2 części, z pacjentami kończącymi udział w Części 1 przed rozpoczęciem leczenia w Części 2. Część 1 badania składa się z 3 równoległych ramion i około 30 pacjentów zostanie jednakowo przydzielonych losowo do jednego z dwóch schematów dawkowania PHA-022121 lub pasujące placebo.
Po zakończeniu Części 1 pacjenci będą kontynuować pojedynczą grupę otwartą w Części 2 badania.
Okres badań przesiewowych wynosi do 8 tygodni, a okresy leczenia wynoszą 12 tygodni (Część 1) i 50 tygodni (Część 2).

- Udokumentowana historia co najmniej 3 ataków HAE w ciągu ostatnich 3 kolejnych miesięcy przed badaniem przesiewowym lub co najmniej 2 ataków HAE w okresie badania przesiewowego
- Niezawodny dostęp i doświadczenie w stosowaniu standardowych leków na ostre napady
Kryteria wyłączenia:
- Ciąża lub karmienie piersią
- Klinicznie istotny nieprawidłowy elektrokardiogram
- Jakakolwiek inna choroba ogólnoustrojowa lub istotna choroba lub zaburzenie, które mogłyby zakłócić bezpieczeństwo pacjenta lub jego zdolność do udziału w badaniu

A new attack was counted if symptoms occurred ≥24 hours after complete resolution of a prior attack.
Attacks were recorded via daily electronic patient-reported outcome (ePRO) diaries and confirmed by the investigator.
Monthly attack rate was calculated as number of attacks during treatment divided by treatment days, multiplied by 28.
Mean rate ratio, confidence interval (CI), and p-value were estimated using a generalized linear model with Poisson distribution, with baseline attack rate included as a factor.
