同种异体干细胞移植 (allo-SCT) 是治疗多种血癌的常用方法。 为确定移植后您的细胞中有多少来自您的供体,医生会完成“嵌合体分析”或对您的细胞进行 DNA 检测。 嵌合体检测有助于医生预测移植物排斥或疾病复发。 NCCC 的医生会定期进行嵌合体检测,通常在移植后 30 到 100 天之间进行。 研究人员认为,在移植后 30 天之内分析嵌合体可能有助于更早地发现问题,让患者更快地接受治疗,并增加移植成功的机会。

我们的嵌合体分析方法依赖于移植前供体和受体的基线 DNA 样本,然后对多态性短串联重复序列 (STR) 进行系列评估,以具体表征当天从我们患者的白细胞中提取的 DNA 的来源(供体与受体) +14/15 异基因干细胞移植后。
首先,从 EDTA 血样的血沉棕黄层中提取 DNA,然后准备使用 STR 标记的 PCR 反应,然后在基因分析仪上评估不同大小的荧光 PCR 产物。
然后可以根据峰高和面积,从分析仪输出中确定对供体和受体嵌合体相对数量的评估。
然后通过细胞分离分离出谨慎的白细胞谱系,并从这些纯化的级分中分离出 DNA 并像全血一样进行评估。
