Cílem této studie je posoudit bezpečnost a vyhodnotit účinnost proti zubnímu kazu nefluoridových zubních pastí COL101 (arginin) ve srovnání s 0,24% fluoridem sodným u dětí ve věku 10-14 let po dobu jednoho roku.

Detailní popis

In this phase 2, double-blind, randomized, active-controlled multi-center study, 2025 subjects were randomized in a ratio of 1:1:1:1 into one of four parallel study groups (1.5% arginine dentifrice, 4.0% arginine dentifrice, 8.0% arginine dentifrice or 0.24% NaF dentifrice with no arginine). Subjects were asked to brush their teeth twice daily at home (unsupervised use) with the assigned dentifrice for a duration of 1 year. Measurements for efficacy, safety and assessment of product use compliance were conducted at baseline, 6 months (±1 month) and 1 year (±1 month). Additional safety evaluations including evaluation of oral soft and hard tissues, as well as product use compliance assessment was performed at 3 months and 9 months (±1 month). All subjects were monitored for adverse events (AEs) during the course of the study.

Kritéria pro zařazení:

  • Do studie budou zařazeni subjekty splňující všechna níže uvedená kritéria:

    1. Souhlas subjektu a informovaný souhlas rodičů/zákonných zástupců s dobrovolnou účastí.
    2. Ochota a schopnost používat přidělené produkty podle pokynů, dostupnost pro všechny schůzky a pravděpodobnost dokončení klinického hodnocení.
    3. Děti ve věku 10-14 let na začátku.
    4. Přítomnost trvalých druhých molárů nebo známky alespoň jedné trvalé erupce druhého moláru, jak je indikováno kupálním průlomem přes sliznici.
    5. Dobrý celkový zdravotní stav doložený přehledem anamnézy.
    6. Subjekty, které mají při vstupu do studie dvě nebo více aktivních kazových lézí (skóre ICDAS 2 nebo vyšší) ve stálých zubech a předchozí zkušenost s kazem (DMFT > 1), D= zkažené, jsou definovány jako skóre ICDAS 3 nebo vyšší

Kritéria vyloučení:

  • Subjekty splňující jakékoli z níže uvedených kritérií budou ze studie vyloučeny:

    1. Přítomnost fixního nebo snímatelného protetického aparátu nebo ortodontické léčby zahrnující více než čtyři stálé zuby.
    2. Užívání léků, které by mohly zvýšit riziko vzniku zubního kazu, tzn.

      léky, které snižují tok slin, a léky s vysokým obsahem cukru.

    3. Dlouhodobá léčba antibiotiky.
    4. Děti s potvrzenou diagnózou kognitivní a/nebo motorické poruchy.
    5. Těžká malokluze.
    6. Subjekty, které při vstupu do studie vykazovaly závažný kaz (ICDAS 5 nebo 6) na pěti nebo více stálých zubech.
    7. Důkaz středně těžkého až těžkého onemocnění parodontu.
    8. Účast na jakékoli jiné klinické studii během 30 dnů před zahájením klinické studie.
    9. Známá historie alergií nebo jiných nežádoucích reakcí na arginin nebo přípravky pro péči o ústní dutinu nebo jejich složky.
    10. Samostatně hlášená anamnéza současného těhotenství, úmyslu otěhotnět během zkušebního období nebo kojení.
