Longitudinale evaluatie van familiale frontotemporale dementie-onderwerpen (LEFFTDS)

Deze multicenter studie zal 300 leden van familiaire Frontotemporale Dementie (FTD) families inschrijven in 8 ervaren FTD onderzoekscentra met een bekende mutatie in MAPT, PGRN of C9ORF72 (100 mutatiedragers met milde dementie of minimaal symptomatische maar niet-demente, 100 asymptomatische mutatie dragers, en 100 klinisch normale familieleden die niet-mutatiedragers zijn) om jaarlijkse beoordelingen te verkrijgen, waaronder T1-MRI, FLAIR, diffusie tensor beeldvorming (DTI), ASL perfusie (ASLp), intrinsieke connectiviteit functionele MRI (icfMRI), MR spectroscopie (MRS ), CSF, bloed en gedrags-, neuropsychologische en functionele beoordeling, voor een totaal van drie beoordelingen per deelnemer.
Een primair doel van deze studie is het identificeren van de meest robuuste en betrouwbare methoden om ziekteprogressie bij familiale FTD te volgen, zodat ziektemodificerende therapeutische onderzoeken op de juiste manier kunnen worden opgezet.

Beschrijving
Inclusiecriteria:
- Moet een familielid zijn met een bekende mutatie in een van de drie belangrijkste FTLD-gerelateerde genen: MAPT, PGRN of C9ORF72.
- Minstens 18 jaar oud.
- Het overheersende fenotype in de verwantschap moet cognitief/gedragsmatig zijn (d.w.z. verwanten waarbij parkinsonisme of ALS het overheersende klinische fenotype is onder getroffen familieleden kunnen worden uitgesloten)
- Zorg voor een betrouwbare informant die minstens wekelijks met dat onderwerp spreekt of hem ziet.
- Onderwerp spreekt voldoende Engels om alle maatregelen te voltooien
- De proefpersoon moet bereid en in staat zijn om in te stemmen met het protocol en moet gedurende 3 jaar jaarlijkse evaluaties ondergaan.
- De proefpersoon moet bereid en in staat zijn om neuropsychologisch onderzoek te ondergaan (in ieder geval bij het basisbezoek).
- De proefpersoon mag geen contra-indicatie hebben voor MRI-beeldvorming.
Uitsluitingscriteria
- Bekende aanwezigheid van een structurele hersenlaesie (bijv. tumor, corticaal infarct).
- Aanwezigheid van een andere neurologische aandoening die bevindingen kan beïnvloeden (bijv. multiple sclerose).
- Proefpersoon is niet bereid om jaarlijks terug te komen voor follow-up, neuropsychologische tests en MR-beeldvorming te ondergaan.
- Onderwerp heeft geen betrouwbare informant.
