Avaliação Longitudinal de Indivíduos com Demência Frontotemporal Familiar (LEFFTDS)

Este estudo multicêntrico incluirá 300 membros de famílias familiares de Demência Frontotemporal (FTD) em 8 centros de pesquisa de FTD experientes com uma mutação conhecida em MAPT, PGRN ou C9ORF72 (100 portadores de mutação com demência leve ou minimamente sintomático, mas não demente, 100 portadores de mutação assintomática portadores e 100 parentes clinicamente normais que não são portadores de mutação) para obter avaliações anuais, incluindo T1-MRI, FLAIR, imagem por tensor de difusão (DTI), perfusão ASL (ASLp), conectividade intrínseca funcional MRI (icfMRI), espectroscopia de RM (MRS ), LCR, sangue e avaliação comportamental, neuropsicológica e funcional, totalizando três avaliações por participante.
Um objetivo principal deste estudo é identificar os métodos mais robustos e confiáveis para rastrear a progressão da doença na FTD familiar, para que os ensaios terapêuticos modificadores da doença possam ser projetados adequadamente.

Descrição
Critério de inclusão:
- Deve ser um membro da família com uma mutação conhecida em um dos três principais genes relacionados ao FTLD: MAPT, PGRN ou C9ORF72.
- Pelo menos 18 anos de idade.
- O fenótipo predominante no parente deve ser cognitivo/comportamental (ou seja, parentes nos quais parkinsonismo ou ELA é o fenótipo clínico predominante entre os parentes afetados podem ser excluídos)
- Tenha um informante confiável que fale pessoalmente ou veja esse assunto pelo menos uma vez por semana.
- O sujeito é suficientemente fluente em inglês para concluir todas as medidas
- O sujeito deve estar disposto e ser capaz de consentir com o protocolo e passar por avaliações anuais ao longo de 3 anos.
- O sujeito deve estar disposto e ser capaz de passar por testes neuropsicológicos (pelo menos na visita inicial).
- O sujeito não deve ter contra-indicação para imagens de ressonância magnética.
Critério de exclusão
- Presença conhecida de uma lesão cerebral estrutural (p. tumor, infarto cortical).
- Presença de outro distúrbio neurológico que possa afetar os achados (por exemplo, esclerose múltipla).
- O sujeito não está disposto a retornar para acompanhamento anual, passar por testes neuropsicológicos e ressonância magnética.
- Sujeito não tem informante confiável.
