Beschreibung
Einschlusskriterien:
Die Probanden müssen alle Einschlusskriterien erfüllen, um an der Studie teilnehmen zu können:
- Erwachsener (>/= 18 Jahre) zum Zeitpunkt des Screenings.
Bestätigte CF-Diagnose basierend auf einem kompatiblen klinischen Syndrom, bestätigt entweder durch einen abnormalen Schweißchloridtest oder zwei CFTR-Genvariationen.*
*Kann aus der Dokumentation in Krankenakten entnommen werden; tatsächliche Testergebnisse nicht erforderlich.
Wahrscheinlich in der Lage, während einer 30-minütigen Sputumsammlung nach einer Behandlung mit hypertoner Kochsalzlösung oder einem anderen Ansatz zur Steigerung der Sputumproduktion mindestens 2 ml Sputum zu produzieren.*
**Durch Urteil des Ermittlers oder seines Beauftragten festgelegt. Ansätze zur Gewinnung von Sputum können inhalierte hypertonische Kochsalzlösung (z. 3 %, 7 % oder 10 %), inhaliertes hypertones Bikarbonat, inhaliertes Mannitol oder spontan ausgespucktes Sputum. Derselbe Ansatz sollte für alle Sputumsammlungen für ein bestimmtes Subjekt verwendet werden.
- P. aeruginosa (unabhängig von koloniebildenden Einheiten (CFU)/ml), das in den letzten 12 Monaten aus einem Sputum, einer Rachenkultur oder einer anderen Atemwegsprobe isoliert wurde.
- Bestätigte Isolierung von P. aeruginosa aus einer Probe des ausgespuckten Sputums beim Screening-Besuch.
- In der Lage, eine informierte Einwilligung zu erteilen.
- In der Lage und bereit, alle Studienbesuche abzuschließen und alle vom Protokoll geforderten Verfahren durchzuführen.
Ausschlusskriterien:
Probanden, die eines der Ausschlusskriterien erfüllen, werden nicht in die Studie aufgenommen:
- Körpergewicht < 30 kg.
- Forciertes Exspirationsvolumen 1 Sekunde < 20 % des vorhergesagten Werts beim Screening unter Verwendung der Hankinson-Gleichungen.
Erhöhte LFTs beim Screening.*

Akute klinische Erkrankung, die ein neues (oral, parenteral) oder inhaliertes(n) Antibiotikum(e) erfordert
*Umfasst keine Medikamente zur chronischen Unterdrückung oder Medikamente mit zyklischer Gabe wie inhalative Antibiotika.
Frauen, die schwanger sind, während des Studienzeitraums eine Schwangerschaft planen oder stillen.* *Frauen im gebärfähigen Alter müssen während des Screenings einen negativen Serum-Beta-Human-Choriongonadotropin-Test haben und sich bereit erklären, für die Dauer der Studie eine wirksame Verhütungsmethode anzuwenden.*
* Eine Frau gilt als gebärfähig, es sei denn, sie ist postmenopausal oder chirurgisch sterilisiert und seit dem Sterilisationsverfahren sind mindestens 3 Monate vergangen.
- Chirurgische Sterilisationsverfahren bei Frauen umfassen Tubenligatur, bilaterale Salpingektomie, Hysterektomie oder bilaterale Oophorektomie.
- Frauen gelten als postmenopausal, wenn sie > 45 Jahre alt sind und mindestens 12 Monate ohne spontane Menstruation ohne andere bekannte oder vermutete Ursache vergangen sind.
- Wirksame Methoden der Empfängnisverhütung umfassen (a) Abstinenz, (b) Partnervasektomie, (c) Intrauterinpessaren, (d) Hormonimplantate (wie Implanon) oder (e) andere hormonelle Methoden (Antibabypille, Injektionen, Pflaster, vaginale Ringe).
- Aktive Behandlung von Mykobakterien oder Pilzorganismen
Voraussichtliche Notwendigkeit, die chronischen Antibiotikatherapien während des Studienzeitraums zu ändern.*
*Probanden, die regelmäßig Medikamente wie inhalative Antibiotika einnehmen, müssen in der Lage sein und ihre Bereitschaft zum Ausdruck bringen, die Therapien zum Zeitpunkt des Screenings für die Dauer der Nachbeobachtungszeit konstant zu halten (entweder auf der Therapie bleiben oder nicht auf der Therapie bleiben) ( ungefähr 30 Tage). Probanden, die eine chronische suppressive antimikrobielle Therapie erhalten, müssen in der Lage sein und ihre Bereitschaft zum Ausdruck bringen, die Therapien für die Dauer ihrer Nachbeobachtungszeit fortzusetzen. Dies schließt eine chronische Azithromycin-Therapie ein.

Number of Co-colonized With Clinically Meaningful Organism Subgroup, Sensitivity Analysis Participants With Desirability of Outcome Ranking (DOOR) Probability Calculated Using the Greatest Reduction by Day 8 Follow-up Visit

A DOOR rank is calculated for each participant based on the following ranking from most (rank 1) to least (rank 4) desirable outcome using each participant's greatest reduction from baseline in log10 P. aeruginosa CFU/mL by Day 8: Rank 1 = No SAE (related to study product) and > 2 log10 reduction in P. aeruginosa CFU/mL. Rank 2 = No SAE (related to study product) and 1-2 log10 reduction in P. aeruginosa CFU/mL. Rank 3 = No SAE (related to study product) and < 1 log10 reduction in P. aeruginosa CFU/mL. Rank 4 = SAE (related to study product). The DOOR probability is estimated using: Pr[DOOR]= Pr[DOORIV > DOORP] + 1/2Pr[DOORIV = DOORP] where DOORIV and DOORP are the DOOR ranks for bacteriophage (phage) and placebo arms, Pr[DOORIV > DOORP] is the proportion of DOOR ranks from IV phage dose exceeding DOOR ranks from placebo, and Pr[DOORIV = DOORP] is the proportion of phage and placebo DOOR ranks being the same. Undetectable colony counts are treated as missing. |
