기본 목표:

I. 재발성/불응성 전신 경쇄(AL) 아밀로이드증에서 이사툭시맙의 확인된 전체 혈액학적 반응률(부분 반응 또는 그 이상)에 의해 측정된 효능을 평가하기 위함.

2차 목표:

I. 이사툭시맙으로 재발성/불응성 AL 아밀로이드증 치료 시 독성을 평가하기 위함.

II. 혈액학적 반응까지의 시간을 평가하기 위해. III. 응답 기간을 평가합니다. IV. 무진행 생존(PFS)을 평가하기 위해. V. 전체 생존(OS)을 평가하기 위해.

3차 목표:

I. 재발성/불응성 면역글로불린 아밀로이드 경쇄(AL) 아밀로이드증에서 장기 특이적 반응률(심장, 신장, 위장관[GI], 간, 연조직, 신경)로 측정한 이사툭시맙의 효능을 장기 반응에 대해 평가할 수 있습니다.

II. 장기 반응에 대해 평가할 수 있는 환자 하위 집합에서 장기 반응까지의 시간을 평가합니다.

개요:

환자는 코스 1의 1일, 8일, 15일 및 22일과 후속 코스의 1일 및 15일에 이사툭시맙을 정맥 주사(IV)받습니다. 치료는 질병 진행 또는 허용할 수 없는 독성이 없는 상태에서 24 과정 동안 28일마다 반복됩니다.

연구 치료 완료 후, 환자는 30일 이내에 추적 관찰되며 이후 최대 4년 동안 최소 6개월마다 추적 관찰됩니다.

  • 환자는 재발성 또는 불응성 원발성 전신 AL 아밀로이드증이 있어야 하며, 편광 현미경의 복굴절에 의한 녹색의 양성 콩고 레드 염색에 의해 조직학적으로 확인되거나, 전자 현미경에 의한 특징적인 외관 및 확증적 AL 아밀로이드 유형(질량 분석 기반 단백체 분석 또는 면역형광)
  • 환자는 등록 전 28일 이내에 측정 가능한 질병이 있어야 합니다. 등록 전 14일 이내에 혈청 베타2 마이크로글로불린, 혈청 정량적 면역글로불린(면역글로불린 [Ig]G, IgA 및 IgM), 혈청 유리 카파 및 람다, M-단백질 정량을 포함한 혈청 단백질 전기영동(SPEP)을 얻어야 합니다.
  • 환자는 등록 전 14일 이내에 관련 혈청 유리 경쇄(카파 또는 람다)와 관련되지 않은 혈청 유리 경쇄의 차이가 >= 4.5mg/dL임을 입증해야 합니다.
